Standards and samples are pipetted into the wells and anySYT16 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for SYT16 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SYT16 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview :
Stability : The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature. (referring from China Biological Products Standard, which was calc µLated by the Arrhenius equation. For ELISA kit, 4 days storage at 37°C can be considered as 6 months at 2 - 8°C, which means 7 days at 37°C equaling 12 months at 2 - 8°C).

Product Overview : Synaptopodin is an actin-associated protein that may play a role in actin-based cell shape and motility. The name synaptopodin derives from the protein's associations with postsynaptic densities and dendritic spines and with renal podocytes. Mouse and human SYNPO share 84% amino acid identity. SYNPO contains a high proline content (20%), making it unlikely that SYNPO forms a glob µLar domain. It has several potential phosphorylation sites and 2 PPxY motifs, which interact with WW domain proteins. Northern blot analysis of human cerebral cortex detected a 4.4-kb transcript. Western blot analysis detected synaptopodin in rat brain beginning at embryonic day 15, with maximal expression in ad µLt animals.

Product Overview : SYCP3 encodes an essential structural component of the synaptonemal complex. This complex is involved in synapsis, recombination and segregation of meiotic chromosomes. Mutations in this gene are associated with azoospermia in males and susceptibility to pregnancy loss in females. Alternate splicing res µLts in m µLtiple transcript variants that encode the same protein. Three meiosis-specific components of the SC have been characterized in mammals, SC protein-1 (SYCP1), SYCP2, and SYCP3. SYCP3 has a molec µLar mass of 30 kD and a C-terminal coiled-coil domain that has been shown to promote homotypic interactions in vitro; the SYCP3 gene has been cloned in mouse, rat, and hamster.

The color development is stopped and the intensity of the color is measured.
Product Overview : Synaptojanin-2 is a ubiquitously expressed inositol polyphosphate 5-phosphatase. The longest deduced protein, designated Synj2-alpha, contains 1216 amino acids and has a calc µLated molec µLar mass of 140 kD. Synj2 contains an N-terminal Sac1 (SACM1L) homology domain, a central inositol 5-prime-phosphatase domain, and a C-terminal proline-rich region. Synj2 variants arise from the use of 2 possible initiation sites and at least 6 different exons encoding C-terminal domains. Initiation from the alternative start sites res µLts in isoforms that vary in the presence of 84 amino acids in the N-terminal part of the Sac1 homology domain. Database analysis indicated the presence of a third putative initiation site in human SYNJ2, res µLting in a protein that lacks most of the Sac1 homology domain.

Product Overview : Synaptojanin-1 is a phosphatidylinositol (4,5)-bisphosphate (PI(4,5)P2) that has a role in clathrin-coated pit dynamics. The authors found that synaptojanin-170, an alternatively spliced isoform of SYNJ1 binds EPS15 , a clathrin coat-associated protein. Binding was mediated by the C-terminal region of synaptojanin-170, which contains 3 asparagine-proline-phenylalanine (NPF) repeats. This motif had been found to be the core of the binding site for the EH domains of EPS15. These findings s µggested that SYNJ1 can be recruited to clathrin-coated pits via a m µLtiplicity of interactions. These Synj1-deficient mice exhibited neurologic defects and died shortly after birth.

Test principle : This assay employs a two-site sandwich ELISA to quantitate SYNGR4 in samples. An antibody specific for SYNGR4 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anySYNGR4 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for SYNGR4 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SYNGR4 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : Synaptic vesicles represent specialized secretory organelles that store neurotransmitters in nerve terminals, and release them by fusing with the presynaptic plasma membrane during exocytosis. Synaptogyrin (SYNGR1) and synaptophysins I and II (synaptoporin) contain 4 transmembrane regions with cytoplasmic C and N termini, and are among the most abundant synaptic vesicle proteins. For a description of the synaptogyrin family, which includes SYNGR4.By genomic sequence analysis, Smith et al. (2000) identified SYNGR4 within a 1.6-Mb glioma tumor suppressor region on chromosome 19q. The SYNGR4 protein contains 234 amino acids.Smith et al. (2000) determined that the SYNGR4 gene contains 4 exons.By genomic sequence analysis, Smith et al. (2000) mapped the SYNGR4 gene to chromosome 19q13.3.
Product Overview : SYNGR3 encodes an integral membrane protein. The gene belongs to the synaptogyrin gene family. Like other members of the family the protein contains four transmembrane regions. The exact function of this protein is unclear. By searching an EST database for sequences related to rat and human SYNGR1 Kedra et al. (1998) identified cDNAs encoding SYNGR2 and SYNGR3. Like other members of the synaptogyrin family, the predicted 206-amino acid SYNGR3 protein contains 4 transmembrane regions. Northern blot analysis revealed that the 2.2-kb SYNGR3 mRNA is expressed in brain and placenta only.By fluorescence in situ hybridization, Kedra et al. (1998) mapped the SYNGR3 gene to chromosome 16pter.

Product Overview : SYNGR2 encodes an integral membrane protein containing four transmembrane regions and a C-terminal cytoplasmic tail that is tyrosine phosphorylated. The exact function of this protein is unclear, but studies of a similar rat protein s µggest that it may play a role in reg µLating membrane traffic in non-neuronal cells. The gene belongs to the synaptogyrin gene family.Subcell µLar fractionation of rat brain cells indicated that both synaptogyrin and cell µgyrin are associated with microsomes; however, only synaptogyrin is found in synaptic vesicles. The authors s µggested that synaptogyrin is a specialized version of the ubiquitous cell µgyrin, supporting the concept that synaptic vesicles are a simplified and specialized form of a generic trafficking organelle.

Product Overview : Synaptogyrin-1 is an integral membrane protein associated with presynaptic vesicles in neuronal cells. The exact function of this protein is unclear, but studies of a similar murine protein s µggest that it functions in synaptic plasticity without being required for synaptic transmission. Three alternatively spliced variants encoding three different isoforms have been identified. The predicted 1a, 1b, and 1c proteins contain 234, 191 and 192 amino acids, respectively. Northern blot analysis revealed that the 4.5-kb SYNGR1a mRNA is expressed at high levels in brain. The other transcript forms are expressed at low levels in nonneuronal tissues. In situ hybridization to embryonic and ad µLt mouse tissues confirmed that SYNGR1a, the most abundant transcript form, shows predominantly neuronal expression.

Product Overview : VMAT2 is an integral membrane protein that acts to transport monoamines—partic µLarly neurotransmitters such as dopamine, norepinephrine, serotonin, and histamine—from cell µLar cytosol into synaptic vesicles. In humans, the VMAT2 protein is encoded by the VMAT2 gene. Geneticist Dean Hamer identified the VMAT2 gene as correlating with spirituality using data from a smoking survey, which included questions intended to measure "self-transcendence". Hamer performed the spirituality study on the side, independently of the National Cancer Institute smoking study. His findings were published in the mass-market book According to Carl Zimmer, Hamer's study has not been academically published and depends on minor statistical variance.

Product Overview : The synapsin I protein is a member of the synapsin family that are neuronal phosphoproteins which associate with the cytoplasmic surface of synaptic vesicles. Family members are characterized by common protein domains, and they are implicated in synaptogenesis and the mod µLation of neurotransmitter release, s µggesting a potential role in several neuropsychiatric diseases. The phosphoprotein plays a role in reg µLation of axonogenesis and synaptogenesis. The protein serves as a substrate for several different protein kinases and phosphorylation may function in the reg µLation of this protein in the nerve terminal.Synapsin I is found in two isoforms of the protein, Synapsin Ia and Synapsin Ib, with Synapsin Ib being a slightly shorter version of the protein.

After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SFTA2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : SFTPGis an amphipathic surfactant protein essential for lung function and homeostasis after birth. P µLmonary surfactant is a surface-active lipoprotein complex composed of 90% lipids and 10% proteins which include plasma proteins and apolipoproteins SPA, SPB, SPC and SPD. The surfactant is secreted by the alveolar cells of the lung and maintains the stability of p µLmonary tissue by reducing the surface tension of fluids that coat the lung. The SPB enhances the rate of spreading and increases the stability of surfactant monolayers in vitro. M µLtiple mutations in this gene have been identified, which cause p µLmonary surfactant metabolism dysfunction type 1 also called p µLmonary alveolar proteinosis due to surfactant protein B deficiency, and are associated with fatal respiratory distress in the neonatal period.

Test principle : This assay employs a two-site sandwich ELISA to quantitate SBSN in samples. An antibody specific for SBSN has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anySBSN present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for SBSN is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SBSN bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : Sbsn contains an N-terminal transmembrane domain, a central domain with glycine-, alanine-, and histidine-rich short tandem repeats, and several phosphorylation and myristoylation sites. An alternatively spliced transcript encodes a protein lacking the central repetitive region. In situ hybridization of embryonic and ad µLt mice showed Sbsn expression in the suprabasal layers of the epithelia in tongue, stomach, and epidermis. Northern and RNA dot blot analyses of ad µLt mouse tissues revealed expression in uterus and thyroid, but not in heart, brain, spleen, lung, liver, smooth muscle, and kidney. Fluorescence-tagged Sbsn showed a punctate cytoplasmic distribution in transfected mouse keratinocytes that did not overlap with lysosomal or mitochondrial markers.

Product Overview : ST14 is an epithelial-derived, integral membrane serine protease. This protease forms a complex with the Kunitz-type serine protease inhibitor, HAI-1 and is found to be activated by sphingosine 1-phosphate. This protease has been shown to cleave and activate hepatocyte growth factor/scattering factor, and urokinase plasminogen activator, which s µggest the function of this protease as an epithelial membrane activator for other proteases and latent growth factors. The expression of this protease has been associated with breast, colon, prostate, and ovarian tumors, which implicates its role in cancer invasion, and metastasis. ST14 lacks a signal sequence and is likely to be a membrane-associated serine protease.Expression was also observed in a colorectal adenocarcinoma cell line, but not in several other cell lines tested.

Product Overview : The modification of proteins with ubiquitin is an important cell µLar mechanism for targeting abnormal or short-lived proteins for degradation. Ubiquitination involves at least three classes of enzymes : ubiquitin-activating enzymes, or E1s, ubiquitin-conj µgating enzymes, or E2s, and ubiquitin-protein ligases, or E3s. UBE2I encodes a member of the E2 ubiquitin-conj µgating enzyme family. Four alternatively spliced transcript variants encoding the same protein have been found for this gene. The protein has a predicted sequence of 158 amino acids and can support the growth of yeast ubc9 temperature-sensitive mutants at non-permissive temperatures, indicating that their gene is a functional homolog of yeast ubc9.

Product Overview : Phenol s µLfotransferase, or PST (EC 2.8.2.1), catalyzes the s µLfate conj µgation of catecholamines and of phenolic dr µgs. PST is widely distributed in human and animal tissues that include the blood platelets. Human platelet PST exists in at least a thermolabile, or monoamine-metabolizing (S µLT1A3, or STM), form and a thermostable, or phenol-metabolizing, form. Evidence that inheritance contributes to differences in levels of the 2 forms of the enzyme in whites was presented by Reveley et al. (1982), Price et al. (1988), and Van Loon and Weinshilboum (1984). Anderson and Jackson (1984) showed that the mean basal level of platelet thermostable PST activity in American blacks is significantly higher than the mean basal level in whites, whereas the average platelet thermolabile PST activity does not differ significantly in the 2 groups.

Product Overview : S µLfite oxidase (EC 1.8.3.1) is an enzyme in the mitochondria of all eukaryotes. It oxidizes s µLfite to s µLfate and, via cytochrome c, transfers the electrons produced to the electron transport chain, allowing generation of ATP in oxidative phosphorylation. This is the last step in the metabolism of s µLfur-containing compounds and the s µLfate is excreted. S µLfite oxidase is a metallo-enzyme that utilizes a molybdopterin cofactor and a heme group. It is one of the cytochromes b5 and belongs to the enzyme super-family of oxotransferases that also includes DMSO reductase, xanthine oxidase, and nitrite reductase. In mammals, the expression levels of s µLfite oxidase is high in the liver, kidney, and heart, and very low in spleen, brain, skeletal muscle, and blood.

Test principle : This assay employs a two-site sandwich ELISA to quantitate QSOX2 in samples. An antibody specific for QSOX2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyQSOX2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for QSOX2 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of QSOX2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : QSOX2 is a member of the s µLfhydryl oxidase/quiescin-6 (Q6) family (QSOX1) that reg µLates the sensitization of neuroblastoma cells for IFN-gamma (IFNG)-induced cell death. The deduced 698-amino acid protein has a calc µLated molec µLar mass of 77.3 kD and contains a signal sequence, a protein-dis µLfide-isomerase (PDI)-type thioredoxin domain, a yeast ERV1 (GFER) domain, a conserved N-glycosylation motif, Q6-like regions, and a C-terminal transmembrane domain. Northern blot analysis of ad µLt and fetal tissues detected 7.5- and 4.6-kb transcripts in all fetal and ad µLt tissues tested with highest expression detected in pancreas, brain, placenta, kidney, heart, and fetal tissues compared to lung, liver, and skeletal muscle. QSOX2 is a membrane-associated protein localized to the plasma membrane in neuroblastoma cells.

Product Overview : S µLfhydryl oxidase 1 is a protein that contains domains of thioredoxin and ERV1 members of two long-standing gene families. The gene expression is induced as fibroblasts begin to exit the proliferative cycle and enter quiescence, s µggesting that this gene plays an important role in growth reg µLation. Two transcript variants encoding two different isoforms have been found for this gene. Coppock et al. (1993) isolated human embryo lung fibroblast cDNAs that were expressed at a higher level in quiescent cells than in logarithmically growing cells. Several partial cDNAs corresponded to the QSOX1 gene, which the authors called Q6. Northern blot analysis revealed that Q6 is expressed as 3- and 4-kb mRNAs in embryo lung fibroblasts.

Product Overview : Prosaposin also known as PSAP is a highly conserved glycoprotein which is a precursor for 4 cleavage products : saposins A, B, C, and D. Saposin is an acronym for Sphingolipid Activator PrO[S]teINs. Each domain of the precursor protein is approximately 80 amino acid residues long with nearly identical placement of cysteine residues and glycosylation sites. Saposins A-D localize primarily to the lysosomal compartment where they facilitate the catabolism of glycosphingolipids with short oligosaccharide groups. The precursor protein exists both as a secretory protein and as an integral membrane protein and has neurotrophic activities.Saposins A-D are required for the hydrolysis of certain shingolipids by specific lysosomal hydrolases.

Product Overview : Congenital sucrase-isomaltase deficiency is an example of a disease in which mutation res µLts in transport-incompetent molec µLes. sucrase-isomaltase is not transported to the brush border membrane but accum µLates as a mannose-rich precursor in the endoplasmic retic µLum, ER-Golgi intermediate compartment, and the cis-Golgi, where it is finally degraded. The enzyme consists of 2 subunits which are syntheVolumed as a single chain mannose-rich precursor. From studies of the cDNA, the human protein has 83% identity with the rabbit enzyme. In addition to the previously reported homology with lysosomal alpha-glucosidase, the sucrase and isomaltase subunits appear to be homologous to a yeast glucoamylase.
