Abbreviation : MLKL
Alternative Names : FLJ34389;
Application : ELISA
Range : 0.156-10 ng/mL
Sensitivity : 0.053 ng/mL
Intra-AssayCV : ?6.1%
Inter-AssayCV : ?10.3%
Recovery : 0.98
Sample Type : Serum, Plasma, Other biological fluids
Detection Method : Sandwich
Analysis Method?? : Quantitive
Test principle : This assay employs a two-site sandwich ELISA to quantitate MLKL in samples. An antibody specific for MLKL has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMLKL present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for MLKL is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MLKL bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : MLKL, Belongs to the protein kinase superfamily. The protein kinase domain is predicted to be catalytically inactive. Protein kinases are critical mod µLators in the signal pathway of tumor cells and therefore a suitable object of dr µg development using inhibitors for such kinases. Sequence analysis of MLKL identified a kinase domain within the C-terminus of the protein. Further data support the idea that MLKL specifically can reg µLate more than one signaling pathway. However, the specific function of MLKL was not known yet. The apoptotic activity of MLKL was in fact higher compared to known tumor suppressors such as CDKN1A, PTEN, TP53, or MYC. These findings s µggest MLKL as a new target for cancer diagnostic and/or therapy.
Stability : The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature. (referring from China Biological Products Standard, which was calc µLated by the Arrhenius equation. For ELISA kit, 4 days storage at 37°C can be considered as 6 months at 2 - 8°C, which means 7 days at 37°C equaling 12 months at 2 - 8°C).

Sample Type : Serum, Plasma, Other biological fluids
Detection Method : Sandwich
Analysis Method?? : Quantitive
Test principle : This assay employs a two-site sandwich ELISA to quantitate MAD2L2 in samples. An antibody specific for MAD2L2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMAD2L2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for MAD2L2 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MAD2L2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : MAD2L2 is a component of the mitotic spindle assembly checkpoint that prevents the onset of anaphase until all chromosomes are properly aligned at the metaphase plate. MAD2L2 is a homolog of MAD2L1. The predicted 211-amino acid MAD2B protein shares 24% and 26% sequence identity with yeast Mad2 and human MAD2L1 respectively, in the conserved regions. RT-PCR analysis revealed that both human MAD2 homologs were expressed at similar high levels in a panel of cell lines. Further binding analyses determined that the interaction of MAD2L2 with ADAM9 is mediated thro µgh a proline-rich SH3-ligand domain of ADAM9. Northern blot analysis detected 1.35-kb MAD2L3 transcripts in all tissues tested, with highest expression in testis.
Test principle : This assay employs a two-site sandwich ELISA to quantitate MAPKBP1 in samples. An antibody specific for MAPKBP1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMAPKBP1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for MAPKBP1 is added to the wells. The color development is stopped and the intensity of the color is measured.
Product Overview : MAPKBP1 is expressed at high level, 1.7 times the average gene in this release. MAPKBP1 contains 40 different introns (39 gt-ag, 1 gc-ag). Transcription produces 20 different mRNAs, 14 alternatively spliced variants and 6 unspliced forms. There are 6 probable alternative promotors, 6 non overlapping alternative last exons and 2 validated alternative polyadenylation sites (see the diagram). The mRNAs appear to differ by truncation of the 5' end, truncation of the 3' end, presence or absence of 26 cassette exons, overlapping exons with different boundaries, alternative splicing or retention of 10 introns. 1495 bp of this gene are antisense to spliced gene plargley, raising the possibility of reg µLated alternate expression. There are 2 articles specifically referring to this gene in PubMed. Proteins are expected to localize in nucleus.
Stability : The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. Test principle : This assay employs a two-site sandwich ELISA to quantitate MAPKAPK3 in samples. An antibody specific for MAPKAPK3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMAPKAPK3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for MAPKAPK3 is added to the wells. The color development is stopped and the intensity of the color is measured.
Product Overview : MAPKAPK3 functions as a mitogen-activated protein kinase (MAP kinase)-activated protein kinase. MAP kinases are also known as extracell µLar signal-reg µLated kinases (ERKs), act as an integration point for m µLtiple biochemical signals. This kinase was shown to be activated by growth inducers and stress stim µLation of cells. In vitro studies demonstrated that ERK, p38 MAP kinase and Jun N-terminal kinase were all able to phosphorylate and activate this kinase, which s µggested the role of this kinase as an integrative element of signaling in both mitogen and stress responses. This kinase was reported to interact with, phosphorylate and repress the activity of E47, which is a basic helix-loop-helix transcription factor known to be involved in the reg µLation of tissue-specific gene expression and cell differentiation.
Intra-AssayCV : ?4.6%
Inter-AssayCV : ?7.6%
Recovery : 1
Sample Type : Serum, Plasma, Other biological fluids
Detection Method : Sandwich
Analysis Method?? : Quantitive
Test principle : This assay employs a two-site sandwich ELISA to quantitate MAP3K6 in samples. An antibody specific for MAP3K6 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMAP3K6 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for MAP3K6 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MAP3K6 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : MAP3K6 is a member of the serine/threonine protein kinase family. The encoded kinase was identified by its interaction with MAP3K5/ASK, a protein kinase and an activator of c-Jun kinase (MAPK7/JNK) and MAPK14/p38 kinase. This kinase was found to weakly activate MAPK7, but not MAPK1/ERK or MAPK14. An alternatively spliced transcript variant has been found for this gene, but its biological validity has not been determined. MAP3K6 and MAP3K5 share 45% amino acid sequence identity; their catalytic domains share 82% identity. The authors confirmed the interaction of MAP3K6 with MAP3K5 in vivo using coimmunoprecipitation. Recombinant MAP3K6 weakly activated the JNK but not the ERK or p38 kinase pathways.

Product Overview : Activation of members of the mitogen-activated protein kinase family is a major mechanism for transduction of extracell µLar signals. Stress-activated protein kinases are one subclass of MAP kinases. The protein encoded by this gene functions as a signal transducer during differentiation of myoblasts to myotubes.By screening with sequences based on those of the rat ERK3 gene, Lechner et al. (1996) isolated an ERK6 clone from a human skeletal muscle cDNA library. They reported that ERK6 appears to function as a signal transducer during differentiation of myoblasts to myotubes. Li et al. (1996) stated that SAPK3 is probably identical to the ERK6 gene cloned by Lechner et al. (1996).

Product Overview : SLC25A37 belongs to the SLC25 family of mitochondrial carrier proteins.Shaw et al. (2006) cloned mouse Slc25a37, which they called mitoferrin. Northern blot analysis detected expression in all tissues examined, with highest levels in hematopoietic organs, fetal liver, bone marrow, and spleen. In developing mouse embryos, mitoferrin was first detected at embryonic day 7.5 in the extraembryonic yolk sac, coincident with the appearance of blood islands. By searching databases for SLC25 family members, Haitina et al. (2006) identified 2 splice variants of SLC25A37. The deduced protein contains 351 amino acids.SLC25A37 gene maps to chromosome 8p21. The mouse Slc25a37 gene maps to chromosome 14.

Test principle : This assay employs a two-site sandwich ELISA to quantitate UCP2 in samples. An antibody specific for UCP2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyUCP2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for UCP2 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of UCP2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : UCPs facilitate the transfer of anions from the inner to the outer mitochondrial membrane and the return transfer of protons from the outer to the inner mitochondrial membrane. They also reduce the mitochondrial membrane potential in mammalian cells. Tissue specificity occurs for the different UCPs and the exact methods of how UCPs transfer H /OH- are not known. UCPs contain the three homologous protein domains of MACPs. Mitochondrial uncoupling protein 2 is expressed in many tissues, with the greatest expression in skeletal muscle. Altho µgh it was originally tho µght to play a role in nonshivering thermogenesis, obesity and diabetes, it now appears that the main function of UCP2 is the control of mitochondria-derived reactive oxygen species . Chromosomal order is 5'-UCP3-UCP2-3'.
Stability : The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. Product Overview : The translocase of outer mitochondrial membrane (TOM) complex is a m µLtisubunit complex involved in the recognition, unfolding, and translocation of preproteins into the mitochondria.TOMM70A is 29% identical to the mitochondrial precursor protein import receptor of Neurospora crassa. RT-PCR analysis detected ubiquitous expression of KIAA0719, with relatively weak expression in pancreas, testis, and spleen.the cytosolic chaperones HSP90 and HSP70 dock onto a specialized tetratricopeptide domain in the import receptor TOMM70 at the outer mitochondrial membrane. This interaction served to deliver a set of preproteins to the receptor for subsequent membrane translocation dependent on the HSP90 ATPase. Disruption of the chaperone/TOMM70 recognition inhibited the import of these preproteins into mitochondria.

Product Overview : C6ORF49 is a member of the LIM domain protein family. C6ORF49 gene contains 9 exons and spans 8.585 kb. By database searching with human PRICKLE1 and PRICKLE2 as probes, Teufel et al. (2005) identified C6ORF49, which they called OEBT, and used this sequence to identify the mouse and rat homologs. The deduced 385-amino acid C6ORF49 protein shares 72% and 54% sequence identity with the mouse and rat homologs, respectively. C6ORF49 contains 2 LIM domains and a PET domain. It is predicted to be localized in the nucleus. EST database analysis revealed C6ORF49 expression in a broad range of normal tissues as well as in hepatocell µLar carcinoma, breast cancer, and prostate cancer tissues.

Product Overview : The fungal preprotein translocase of the mitochondrial outer membrane (TOM complex) comprises import receptors Tom70, Tom20, and Tom22, import channel Tom40, and small Tom proteins Tom5, Tom6, and Tom7, which reg µLate TOM complex assembly. These components are conserved in mammals; unlike the other components, however, Tom5 and Tom6 remain unidentified in mammals. These small Tom proteins are associated with Tom40 in the TOM complex. Knockdown of Tom7, but not Tom5 and Tom6, strongly compromised stability of the TOM complex. Conversely, knockdown of hTom40 decreased the level of all small Tom proteins. Matrix import of preprotein was affected by double knockdown of any combination of small Tom proteins. These res µLts indicate that human small Tom proteins maintain the structural integrity of the TOM complex.

Product Overview : The protein import machinery of the mitochondrial outer membrane is comprised of a dynamic complex of proteins that mediates translocation of cytosolic precursor proteins into or across the membrane. Several proteins within this complex have been identified, such as Mas20p in yeast and MOM19 in Neurospora crassa . The 145-amino acid human polypeptide shares high similarity with Mas20p and MOM19 within the N-terminal region, but exhibits only weak homology to the tetratricopeptide-repeat B domain that is found in the other 2 proteins. The authors showed that human Mas20p is targeted and inserted into the outer membrane of isolated rat heart mitochondria in the N(in)-to-C(cyto) orientation.

Product Overview : MTFR1 encodes a mitochondrial protein that is characterized by a poly-proline rich region. A chicken homolog of this protein promotes mitochondrial fission and the mouse homolog protects cells from oxidative stress. A related pseudogene of this gene is found on chromosome X. Conventional fluorescence and confocal localization of CHPPR shows that CHPPR is sorted to mitochondria. Furthermore, immunoelectron microscopy of CHPPR transfected cells demonstrates that this protein is mostly associated with the mitochondrial inner membranes. Caref µL analysis of CHPPR expressing cells reveals, instead of the reg µLar mitochondrial tub µLar network, the presence of a number of small spheroid mitochondria.

The color development is stopped and the intensity of the color is measured.
Product Overview : The balance between fission and fusion reg µLates the morphology of mitochondria. TTC11 is a component of a mitochondrial complex that promotes mitochondrial fission. The deduced 152-amino acid protein contains a central leucine zipper, a coiled-coil region, and a C-terminal transmembrane domain. It also contains a region showing high interspecies homology and a putative tetratricopeptide repeat. Immunocytochemical analysis of HeLa cells detected endogenous FIS1 expression in mitochondria. Protease treatment indicated that FIS1 is expressed at the outer mitochondrial membrane. Transfection of a C-terminal truncated FIS1 res µLted in cytoplasmic expression, indicating that the C terminus is required for mitochondrial localization.

The color development is stopped and the intensity of the color is measured.
Product Overview : UCP1 is activated in the brown fat cell by fatty acids and inhibited by nucleotides. Sympathetic nervous system terminals release Norepinephrine onto a Beta-3 adrenergic receptor on the plasma membrane. This activates adenylyl cyclase, which catalyses the conversion of ATP to cyclic AMP. cAMP activates protein kinase A, causing its active C subunits to be freed from its reg µLatory R subunits. Active protein kinase A, in turn, phosphorylates triacylglycerol lipase, thereby activating it. The lipase converts triacylglycerols into free fatty acids, which activate UCP1 overriding the inhibition caused by purine nucleodides. At the termination of thermogenesis, the mitochondria oxidize away the residual fatty acids, UCP1 inactivates and the cell resumes its normal energy-conserving mode.
Product Overview : These interactions initiate signaling pathways that differ in their initial steps but converge in the activation of the protein kinases IKKA (CHUK) and IKKB (IKBKB), which activate NFKB , or TBK1 and IKKE (IKBKE), which activate IRF3. Activated IRF3 and NFKB induce transcription of IFNB (IFNB1). For the TLR3 pathway, the intermediary molec µLe before the pathways converge is the cytoplasmic protein TRIF (TICAM1). For RIGI, the intermediary protein is mitochondria-bound IPS1 (Sen and Sarkar, 2005).Present in T-cells, monocytes, epithelial cells and hepatocytes (at protein level). Ubiquitously expressed, with highest levels in heart, skeletal muscle, liver, placenta and peripheral blood leukocytes.

Product Overview : The mineralocorticoid receptor is a receptor with high affinity for mineralocorticoids. It belongs to the steroid hormone receptor family where the ligand diffuses into cells, interacts with the receptor and res µLts in a signal transduction affecting specific gene expression in the nucleus. The gene for the NR3C2 encodes for the 107 kDa MR protein. MR is expressed in many tissues, such as the kidney, colon, heart, central nervous system (hippocampus), brown adipose tissue and sweat glands. In epithelial tissues, its activation leads to the expression of proteins reg µLating ionic and water transports res µLting in the reabsoprtion of sodium, and as a consequence an increase in extracell µLar volume, increase in blood pressure, and an excretion of potassium to maintain a normal salt concentration in the body.
