Product Overview : DLL4 is a homolog of the Drosophila delta gene. The delta gene family encodes Notch ligands that are characterized by a DSL domain, EGF repeats, and a transmembrane domain. The predicted 685-amino acid DLL4 protein exhibits all of the hallmarks of the Delta family of Notch ligands. It has an extracell µLar region containing 8 EGF-like repeats and a DSL domain involved in Notch binding, as well as a transmembrane domain and a cytoplasmic tail that lacks catalytic motifs. Human DLL4 shares 87% amino acid sequence identity with mouse Dll4. Northern blot analysis of mouse tissues detected highest Dll4 expression in lung, followed by heart, kidney, skeletal muscle, and brain; Dll4 expression was barely detectable in spleen and testis.

Product Overview : DLL1 is a human homolog of the Notch Delta ligand and is a member of the delta/serrate/jagged family. It plays a role in mediating cell fate decisions during hematopoiesis. It may play a role in cell-to-cell communication. Sequence analysis predicted that the 723-amino acid DLL1 transmembrane protein, which is 88% identical to the mouse Dll1 protein, has a DSL domain followed by 8 tandem EGF-like repeats and a short cytoplasmic C-terminal region. Northern blot analysis revealed strongest expression of 4.0- and 4.6-kb transcripts in heart and pancreas, with lower expression in brain and muscle and almost no expression in placenta, lung, liver, and kidney. Test principle : This assay employs a two-site sandwich ELISA to quantitate DMBT1 in samples. An antibody specific for DMBT1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyDMBT1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for DMBT1 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of DMBT1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : Loss of sequences from human chromosome 10q has been associated with the progression of human cancers. The gene DMBT1 was originally isolated based on its deletion in a med µLloblastoma cell line. DMBT1 is expressed with transcripts of 6.0, 7.5, and 8.0 kb in fetal lung and with one transcript of 8.0 kb in ad µLt lung, altho µgh the 7.5 kb transcript has not been characterized. The DMBT1 protein is a glycoprotein containing m µLtiple scavenger receptor cysteine-rich (SRCR) domains separated by SRCR-interspersed domains (SID). Transcript variant 2 (8.0 kb) has been shown to bind surfactant protein D independently of carbohydrate recognition. This indicates that DMBT1 may not be a classical tumor supressor gene, but rather play a role in the interaction of tumor cells and the immune system.

Product Overview : DLC1 contains 37 exons and spans approximately 59 kb. The predicted DLC1 protein contains 1755 amino acids. Expression of DLC1 significantly suppressed the growth of 4 different cancer cell lines. Northern blot analysis revealed that DLC1 was expressed as 6- and 8-kb mRNAs in all tissues tested, most abundantly in prostate and testis. the 8-kb mRNA and several other alternatively spliced DLC1 transcripts contain disrupted coding regions and are likely to encode nonfunctional proteins. RT-PCR experiments indicated that 33% of primary lung, esophageal, and renal cancers and cancer cell lines tested either lacked DLC1 transcripts entirely or contained increased levels of nonfunctional DLC1 mRNA. The authors concluded that aberrant transcription of DLC1 may be involved in carcinogenesis of the lung, esophagus, and kidney.
