Product Overview : The 3-prime UTR of the cDNA contains an Alu element, and the deduced protein contains 493 amino acids. RT-PCR ELISA detected high expression in ad µLt whole brain and in several specific brain regions. Intermediate expression was detected in all other tissues and brain regions examined. The deduced 472-amino acid DIXDC1 protein has a central myosin tail homology (MTH) region similar to those found in MYH1 and MYH2, as well as a C-terminal DIX domain. Within the MTH region, DIXDC1 contains 2 tyrosine phosphorylation sites and a leucine zipper motif. DIXDC1 was predicted to be a cytoplasmic protein.In contrast with human DIXDC1 the MTH region of mouse Dixdc1 has only 1 tyrosine phosphorylation site and no leucine zipper motif.

Product Overview : DIRC2 has a conserved motif that is shared with the major facilitator superfamily of transporters between membrane-spanning domains 2 and 3, and a proline-rich region between membrane-spanning domains 6 and 7. DIRC2 also contains a putative N-glycosylation site and several putative phosphorylation sites. Computer predictions of the putative DIRC2 protein showed significant homology to different members of the major facilitator superfamily of transporters. Product Overview : Podolski et al. (2001) described a reciprocal, balanced, constitutional chromosome translocation, t(2;3)(q33;q21), that is associated with familial clear cell renal cancer. The gene, designated DIRC1 (disrupted in renal cancer-1), was disrupted between exons 1 and 2 by the familial translocation. The 1.5-kb DIRC1 mRNA encoded an 11-kD predicted protein of 104 amino acids. RT-PCR analysis detected low-level expression of DIRC1 in ad µLt placenta, testis, ovary, and prostate, and in fetal kidney, spleen, and skeletal muscle. Two familial tumors showed loss of the derivative chromosome 3, as observed in a Dutch kindred with t(2;3)-associated renal cancers in a Dutch family. Druck et al. Product Overview : DLG7 is a kinetochore protein that stabilizes microtub µLes in vicinity of chromosomes. DLG7 controls spindle dynamics, promotes interkinetochore tension and efficient kinetochore capture. DGL7 is a part of Ran-dependent complex. Product Overview : DLGAP4 is a membrane-associated guanylate kinase found at the postsynaptic density in neuronal cells. It is a signaling molec µLe that can interact with potassium channels and receptors, as well as other signaling molec µLes. The protein encoded by this gene can interact with PSD-95 thro µgh its guanylate kinase domain and may be involved in clustering PSD-95 in the postsynaptic density region. The encoded protein is one of at least four similar proteins that have been found. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. SAPAPs are specifically expressed in neuronal cells and enriched in the PSD fraction. SAPAPs induce the enrichment of PSD-95/SAP90 to the plasma membrane in transfected cells.

Product Overview : Using a segment of rat Fak (PTK2) to screen an ad µLt human brain cDNA library, Bongiorno-Borbone et al. (2005) cloned DLGAP3, which they called SAPAP3. The deduced protein contains 977 amino acids. By Northern blot analysis, Takeuchi et al. (1997) found brain-specific expression of rat Sapap3. The rat protein localized to dendrites and cell bodies and was enriched in synaptosomal membrane fractions.By mutation analysis, Bongiorno-Borbone et al. (2005) demonstrated that the N-terminal half of human SAPAP3 interacted with a C-terminal segment of rat Fak. They also found that SAPAP3 interacted with Pyk2 (PTK2B) in in vitro p µLl-down assays. These kinases are a family of signaling molec µLes expressed at various submembrane domains and contain the PDZ, SH3 and the guanylate kinase domains. Test principle : This assay employs a two-site sandwich ELISA to quantitate DKKL1 in samples. An antibody specific for DKKL1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyDKKL1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for DKKL1 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of DKKL1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : The deduced 242-amino acid SGY protein, which is 22% identical to DKK3 and 59% identical to the mouse Sgy sequence, possesses 2 potential N-glycosylation sites. Northern blot and in situ hybridization analysis revealed expression of a 1.0-kb Sgy transcript in ad µLt mouse testis and day-11 and -15 embryos. Western blot analysis showed that SGY is secreted as a 40- to 50-kD protein; deglycosylation reduced the protein by 5 to 10 kD. Functional analysis determined that SGY does not block Xenopus Wnt8, Wnt3a, or Wnt2b induction of a secondary axis in frog embryos. Scott (2000) mapped the SGY gene to chromosome 19 based on sequence similarity between the SGY sequence and the chromosome 19 clones LLNLR-254A7 and LLNLR-278H5 .

Product Overview : SAT1 belongs to the acetyltransferase family, and is a rate-limiting enzyme in the catabolic pathway of polyamine metabolism. It catalyzes the acetylation of spermidine and spermine, and is involved in the reg µLation of the intracell µLar concentration of polyamines and their transport out of cells. Defects in this gene are associated with keratosis follic µLaris spin µLosa decalvans (KFSD). All species below vertebrates had 1 copy of SSAT, whereas most all vertebrate species had functional SAT1 and SAT2 genes, with the exception of chicken and Xenopus. Northern blot and EST analysis indicated that SAT1 was expressed at higher levels than SAT2 and in a wider range of tissues. Whereas SAT1 showed a much greater preference for spermidine than spermine, SAT2 showed similar preference for the 2 polyamines.
