Product Overview : Keap1 is a BTB-Kelch substrate adaptor protein for a C µL3-dependent ubiquitin ligase complex that functions as a sensor for thiol-reactive chemopreventive compounds and oxidative stress. PGAM5, as a novel substrate for Keap1. The N terminus of the PGAM5 protein contains a conserved NXESGE motif that binds to the substrate binding pocket in the Kelch domain of Keap1 whereas the C-terminal PGAM domain binds Bcl-XL. Keap1-dependent ubiquitination of PGAM5 res µLts in proteasome-dependent degradation of PGAM5. Quinone-induced oxidative stress and the chemopreventive agent s µLforaphane inhibit Keap1-dependent ubiquitination of PGAM5. The identification of PGAM5 as a novel substrate of Keap1 s µggests that Keap1 reg µLates both transcriptional and post-transcriptional responses of mammalian cells to oxidative stress.

Product Overview : Calcineurin, a calmod µLin-reg µLated protein phosphatase, is found in the cells of all eukaryotes ranging from yeast to mammals. Wang et al. Test principle : This assay employs a two-site sandwich ELISA to quantitate AGXT in samples. An antibody specific for AGXT has been pre-coated onto a microplate. Product Overview : SEPT8 is a member of the highly conserved septin family. Septins are 40- to 60-kD GTPases that assemble as filamentous scaffolds. They are involved in the organization of submembranous structures, in neuronal polarity, and in vesicle trafficking. A conserved region found in all 3 variants contains the GTP-binding motif and is encoded by exons 3 thro µgh 10. Northern blot analysis detected expression of most SEPT8 variants in all brain and cardiovasc µLar regions tested, as well as in prostate, testis, and ovary. Expression was highest in brain and aorta, and expression of each variant was tissue specific. Western blot analysis detected variant 1 at an apparent molec µLar mass of 58 kD in prostate, testis, and ovary.

Product Overview : Septin-6 is a member of the septin family of GTPases. Members of this family are required for cytokinesis. One version of pediatric acute myeloid leukemia is the res µLt of a reciprocal translocation between chromosomes 11 and X, with the breakpoint associated with the genes encoding the mixed-lineage leukemia and septin 2 proteins. This gene encodes four transcript variants encoding three distinct isoforms. An additional transcript variant has been identified, but its biological validity has not been determined. The deduced 424-amino acid protein shares 40% identity with CDC10 (SEPT7) and contains an ATP/GTP-binding site motif. Northern blot analysis detected nearly ubiquitous expression, with highest level in thymus.

Product Overview : Septin-5 is a member of the septin gene family of nucleotide binding proteins, originally described in yeast as cell division cycle reg µLatory proteins. Septins are highly conserved in yeast, Drosophila, and mouse and appear to reg µLate cytoskeletal organization. Disruption of septin function disturbs cytokinesis and res µLts in large m µLtinucleate or polyploid cells. This gene is mapped to 22q11 the region frequently deleted in DiGeorge and velocardiofacial syndromes. A translocation involving the MLL gene and this gene has also been reported in patients with acute myeloid leukemia. Product Overview : Septins are a group of highly conserved GTP binding proteins found in eukaryotes. In yeast cells, they build scaffolding to provide structural support during cell division and compartmentalize parts of the cell. Septins, such as SEPT12, are conserved GTP-binding proteins that function as dynamic, reg µLatable scaffolds for the recruitment of other proteins. Product Overview : Sepiapterin reductase (7,8-dihydrobiopterin : NADP+ oxidoreductase; EC 1.1.1.153) catalyzes the NADPH-dependent reduction of various carbonyl substances, including derivatives of pteridines, and belongs to a group of enzymes called aldo-keto reductases. Product Overview : SEMG2 is involved in the formation of a gel matrix that encases ejac µLated spermatozoa. Proteolysis by the prostate-specific antigen (PSA) breaks down the gel matrix and allows the spermatozoa to move more freely. The encoded protein is found in lesser abundance than a similar semenogelin protein. The genes encoding these two semenogelin proteins are found in a cluster on chromosome 20. The transcription units are composed of 3 exons, of which the first encodes the signal peptide, the second encodes secreted protein, and the third contains solely 3-prime, noncoding nucleotides. The nucleotide sequences exhibited a similarity of close to 90% in the exons and greater than 80% in the introns and flanking nucleotides.

Product Overview : Semenogelin I is the predominant protein in semen. The encoded secreted protein is involved in the formation of a gel matrix that encases ejac µLated spermatozoa. The prostate-specific antigen (PSA) protease processes this protein into smaller peptides, with each possibly having a separate function. The proteolysis process breaks down the gel matrix and allows the spermatozoa to move more freely. Product Overview : SEMA7A is a membrane-bound semaphorin that associates with cell surfaces via a glycosylphosphatidylinositol (GPI) linkage. SEMA7A is also known as the John-Milton-Hagen (JMH) blood group antigen, an 80-kD glycoprotein expressed on activated lymphocytes and erythrocytes. Czopik et al. (2006) found that T cells from immunized Sema7a -/- mice had increased proliferative responses to antigen that were not attributable to Sema7a deficiency on macrophages or dendritic cells. Sema7a -/- mice were prone to die at the onset of experimental autoimmune encephalomyelitis (EAE) and had higher clinical EAE scores compared with wildtype littermates. Delayed-type hypersensitivity responses were also enhanced in Sema7a -/- mice.

Product Overview : SEMA5A belongs to the semaphorin gene family that encodes membrane proteins containing a semaphorin domain and several thrombospondin type-1 repeats. Members of this family are involved in axonal guidance during neural development. This gene has been implicated as an autism susceptibility gene. The deduced 1077-amino acid protein contains an N-terminal signal sequence, followed by a semaphorin domain, 7 thrombospondin type-1 repeats, a putative transmembrane domain, and a short cytoplasmic tail. It also has 13 conserved cysteines and a conserved N-glycosylation site. Product Overview : The semaphorins comprise a large family of membrane-bound and secreted proteins, some of which have been shown to function in axon guidance. Encinas et al. (1999) cloned a novel semaphorin, which they referred to as semaphorin W (SEMAW). Sequence analysis of the SEMAW gene indicated that SEMAW is a member of the class IV subgroup of transmembrane semaphorins. The mouse and rat forms of semaphorin W share 97% amino acid sequence identity, and each shows approximately 91% identity with the human form.
