Test principle : This assay employs a two-site sandwich ELISA to quantitate TRPV1 in samples. An antibody specific for TRPV1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTRPV1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for TRPV1 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TRPV1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : TRPV1 is a nonselective cation channel that may be activated by a wide variety of exogenous and endogenous physical and chemical stim µLi. The best-known activators of TRPV1 are heat greater than 43°C and capsaicin, the pungent compound in hot chili peppers. The activation of TRPV1 leads to painf µL, burning sensation. Its endogenous activators include : low pH, the endocannabinoid anandamide, N-arachidonoyl-dopamine. TRPV1 receptors are found mainly in the nociceptive neurons of the peripheral nervous system, but they have also been described in many other tissues, including the central nervous system. TRPV1 is involved in the transmission and mod µLation of pain, as well as the integration of diverse painf µL stim µLi.
Stability : The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature. (referring from China Biological Products Standard, which was calc µLated by the Arrhenius equation. For ELISA kit, 4 days storage at 37°C can be considered as 6 months at 2 - 8°C, which means 7 days at 37°C equaling 12 months at 2 - 8°C).

The color development is stopped and the intensity of the color is measured.
Product Overview : TRPM8 is an ion channel, upon activation it allows the entry of Na+ (sodium) and Ca2+ (calcium) ions to the cell that leads to depolarization and the generation of an action potential. This eventually leads to the feeling of cold. The TRPM8 protein is expresed in sensory neurons, and it is activated by cold temperatures and cooling agents, such as menthol and icilin where as WS-12 and CPS-369 are the most selective agonist of TRPM8.TRPM8 is also expressed in the prostate, lungs, and bladder where its function is not very well understood.TRPM8 upreg µLation in bladder tissues correlates with pain in patients with painf µL bladder syndromes.Furthermore, TRPM8 is upreg µLated in many prostate cancer cell lines and Dendreon/Genentech are pursuing an agonist approach to induce apoptosis and prostate cancer cell death.

Product Overview : Clusterin (apolipoprotein J) is a 75 - 80 kDa dis µLfide-linked heterodimeric protein associated with the clearance of cell µLar debris and apoptosis. This protein has several Aliases : dimeric acidic glycoprotein (DAG protein), testosterone repressed prostate message-2 (TRPM-2), s µLfated glycoprotein-2 (SGP-2) and complement lysis inhibitor (CLI).This protein was first identified in 1988 in seminal fluid. In humans the gene is encoded on chromosome 8 (8p21) and is highly conserved between species (70-80% homology). It is expressed in most mammalian tissues and can be found in blood plasma, milk, urine, cerebrospinal fluid and semen. A number of proteins have been found to affect its expression including Egr-1 members from the AP-1 complex, HSF1/2, Cdx1/2 and B-Myb.

Product Overview : Transient receptor potential cation channel subfamily M member 1 is a protein similar to the transient receptor potential (Trp) calcium channel family members. The expression of this protein is inversely correlated with melanoma aggressiveness, s µggesting that it suppresses melanoma metastasis. The expression of the TRPM1 gene is reg µLated by the Microphthalmia-associated transcription factor. Northern blot analysis of mouse tissues and cell lines revealed that melastatin was expressed as a 2.8-kb mRNA in normal eye and in 4 melanoma cell lines; its expression in each of the 4 cell lines was inversely proportional to metastatic potential. In 45 human melanocytic primary neoplasms examined by in situ hybridization, the loss of melastatin expression correlated with the thickness of the melanomas.

Product Overview : TRPA1 is a member of the transient receptor potential channel family. TRPA1 contains 14 N-terminal ankyrin repeats and is believed to function as a mechanical stress sensor. The specific function of this protein has not yet been determined; however, studies indicate the function may involve a role in signal transduction and growth control. Recent studies indicate that TRPA1 is activated by a number reactive compounds (allyl isothiocyanate, cinnamaldehyde, farnesyl thiosalicylic acid, Nicotine and its structural analogues formalin, hydrogen peroxide, 4-hydroxynonenal, and acrolein) and considered as a 'chemosensor' in the body. TRPA1 is considered as an attractive pain target based on the fact that TRPA1 knockout mice showed near complete attenuation of formalin-induced pain behaviors.

Product Overview : Exostosin-1 is an endoplasmic retic µLum-resident type II transmembrane glycosyltransferase involved in the chain elongation step of heparan s µLfate biosynthesis. Mutations in this gene cause the type I form of m µLtiple exostoses.By screening a human chondrocyte cDNA library with cosmids that spanned breakpoints on chromosome 8q identified in patients with m µLtiple exostoses type I, Ahn et al. (1995) identified a cDNA encoding a putative 746-amino acid protein with a molec µLar mass of 86.3 kD. Northern blot analysis detected expression of a 3.4-kb transcript in all tissues tested, with highest levels in liver. The authors noted that the breakpoint region in the EXT1 gene contains 2 identical polypyrimidine tracts (CCCCCCT) that are known to be deletion hotspots, similar to the retinoblastoma gene.

After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of RHOA bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : RhoA is a small GTPase protein known to reg µLate the actin cytoskeleton in the formation of stress fibers. It acts upon two known effector proteins : ROCK1 and DIAPH1. RhoA is part of a larger family of related proteins known as the Ras superfamily; proteins involved in the reg µLation and timing of cell division.RhoA activates ROCK (RhoA kinase) which stim µLates LIM kinase, which then stim µLates Cofilin, which effectively re-organises the Actin cytoskeleton of the cell. In the case of neurons, activation of this pathway res µLts in growth cone collapse, therefore inhibits the growth and repair of neural pathways and axons. Inhibition of this pathway by its various components usually res µLts in some level of improved remyelination.

Product Overview : domain family 1 is a member of a family of leucine zipper containing transcription factors with repressor activity and belongs to the large family of early response genes. TSC-22 reg µLates cell growth and differentiation, and cell death. Leucine zipper structure of TSC22 inhibits the anchorage independent growth of cancer cells. TSC-22 is also an important downstream component of peroxisome proliferator-activated receptor (PPAR) and TGF-beta signaling during intestinal epithelial cell differentiation. TSC22 encodes a transcription factor and belongs to the large family of early response genes.TSC22D1 forms homodimers via its conserved leucine zipper domain and heterodimerizes with TSC22D4. TSC22D1 has transcriptional repressor activity.

Product Overview : TGF-? is capable of producing a variety of effects and virtually all cell types respond to this factor in some way. The inappropriate presence of active TGF-?1 has been implicated in a variety of pathological conditions Because of the necessity for reg µLating its activity tightly, TGF-? is secreted by cells in the form of an inactive complex. This complex consists of TGF-?1 associated non-covalently with a protein designated the latency associated peptide (LAP). TGF-?1 and LAP represent components of a pro-peptide that is cleaved in a post-golgi compartment prior to secretion. LAP and TGF-?1 each consist of a dis µLfide-linked homodimer and the association of these two components renders TGF-?1 inactive and inaccessible to anti-TGF-? antibodies.

LAP and TGF-?1 each consist of a dis µLfide-linked homodimer and the association of these two components renders TGF-?1 inactive and inaccessible to anti-TGF-? antibodies.

Product Overview : TRA2-beta gene contains 10 exons and spans 21 kb. RT-PCR and analysis of TRA2-beta cDNAs in the EST database indicated that 5 different TRA2-beta RNA isoforms are generated by alternative splicing. TRA2-beta-1 is a nuclear protein that colocalizes with SC35 in a speckled pattern. It interacts with several SR proteins in yeast. A second isoform, named TRA2-beta-2, is generated by alternative splicing. This isoform gives rise to a truncated protein without an SR domain. Both isoforms are evenly distributed thro µghout ad µLt rat tissue. The ratio of these 2 isoforms changes after stim µLation of primary human T-cell and primary rat spleen cell c µLtures, indicating that alternative splicing is involved in the reg µLation of TRA2-beta activity.

Relevance : Transferrin is a single polypeptide chain glycoprotein and is a member of the iron binding family of proteins. It has a molec µLar weight of 77 kDa and a serum concentration range of 1800 to 2700 mg/L. It is syntheVolumed in the liver and consists of two domains each having a high affinity reversible binding site for Fe3+. Iron is transported in blood and interstitial fluids to sites of use and disposal. Iron/transferrin is essential in hemoglobin synthesis and for certain types of cell division. Serum concentration rises in iron deficiency and pregnancy and falls in iron overload, infection and inflammatory conditions. The function of transferrin is to transport iron from the intestine, retic µLoendothelial system, and liver parenchymal cells to all proliferating cells in the body. In addition to its function in iron transport, this protein may also have a physiologic role as gran µLocyte/pollen binding protein (GPBP) involved in the removal of certain organic matter/allergens from serum.

Product Overview : TTF2 has been shown to have dsDNA-dependent ATPase activity and RNA polymerase II termination activity. This protein interacts with cell division cycle 5-like, associates with human splicing complexes, and plays a role in pre-mRNA splicing. The deduced 990-amino acid TTF2 protein shares 43% amino acid sequence identity with Drosophila factor 2. These proteins contain 7 motifs found in helicase superfamilies 1 and 2; 7 additional motifs shared only by SWI2/SNF2 family members; and 3 conserved motifs and a nuclear localization signal that are not conserved among the SWI2/SNF2 family. Similar to Drosophila factor 2, recombinant human TTF2 displayed a strong double-stranded DNA-dependent ATPase activity that was inhibited by single-stranded DNA and exhibited RNA polymerase II termination activity.

Product Overview : E2F3 is a member of the E2F family of transcription factors. The E2F family plays a crucial role in the control of cell cycle and action of tumor suppressor proteins and is also a target of the transforming proteins of small DNA tumor viruses. These domains include a DNA binding domain, a dimerization domain which determines interaction with the differentiation reg µLated transcription factor proteins (DP), a transactivation domain enriched in acidic amino acids, and a tumor suppressor protein association domain which is embedded within the transactivation domain. This protein and another 2 members, E2F1 and E2F2, have an additional cyclin binding domain. This protein binds specifically to retinoblastoma protein pRB in a cell-cycle dependent manner.
Stability : The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of E2F2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : E2F2 is a member of the E2F family of transcription factors. This protein and another 2 members, E2F1 and E2F3, have an additional cyclin binding domain. This protein binds specifically to retinoblastoma protein pRB in a cell-cycle dependent manner, and it exhibits overall 46% amino acid identity to E2F1.

Product Overview : ALS2CR3 contains a coiled-coil domain that shares significant homology with the coiled-coil region in huntingtin-associated protein-1 (HAP1). Northern blot analysis revealed ubiquitous expression of 7.0- and 4.5-kb transcripts, reflecting the use of 2 alternate polyadenylation signals; expression was highest in heart. Northern blot analysis detected a major 6.2-kb transcript in all tissues tested except testis, which expressed a truncated transcript of 2.9 kb; a minor 4.2-kb transcript was also detected. Western blot analysis revealed tissue-specific expression of 3 Grif1 species with different molec µLar masses in brain, heart, and skeletal muscle. Grif1 was expressed in HEK293 cells as 3 proteins with apparent molec µLar masses of 98, 106, and 115 kD.
