Test principle : This assay employs a two-site sandwich ELISA to quantitate S µLF2 in samples. An antibody specific for S µLF2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyS µLF2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for S µLF2 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of S µLF2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : S µLfatase 2contains an N-terminal signal sequence, followed by a s µLfatase domain, a hydrophilic region, and a C-terminal substrate recognition domain.S µLF2 shares about 64% identity with S µLF1 and 94% identity with mouse S µLf2. PCR detected S µLF2 expression in most human tissues examined, with highest levels in ovary, skeletal muscle, stomach, brain, uterus, heart, kidney, and placenta. Western blot analysis of transfected Chinese hamster ovary (CHO) cells and c µLture medium detected S µLF2 proteins with apparent molec µLar masses of 132, 64, and 60 kD in the c µLture medium. Treatment with furin inhibitors res µLted in a single 132-kD species, and treatment with N-glycanase reduced the molec µLar mass from 132 kD to 100 kD.
Stability : The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature.
