Test principle : This assay employs a two-site sandwich ELISA to quantitate VD3 in samples. An antibody specific for VD3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyVD3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for VD3 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of VD3 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : Cholecalciferol is a form of vitamin D, also called vitamin D3. It is structurally similar to steroids such as testosterone, cholesterol, and cortisol (tho µgh vitamin D3 itself is a secosteroid). Cholecalciferol is produced industrially for use in vitamin supplements and to fortify foods by the µLtraviolet irradiation of 7-dehydrocholesterol extracted from lanolin found in sheep's wool. Paraphrasing a more detailed explanation, cholesterol is extracted from wool grease and wool wax alcohols obtained from the cleaning of wool after shearing. The cholesterol undergoes a four step process to make 7-dehydrocholesterol, the same compound that is stored in the skin of animals. The 7-dehydrocholesterol is then irradiated with µLtra violet light. Rodents are somewhat more susceptible to high doses than other species, and cholecalciferol has been used in poison bait for the control of these pests.

Product Overview : ADAMTS4 encodes a member of the ADAMTS (a disintegrin and metalloproteinase with thrombospondin motifs) protein family. Members of the family share several distinct protein mod µLes, including a propeptide region, a metalloproteinase domain, a disintegrin-like domain, and a thrombospondin type 1 (TS) motif. Individual members of this family differ in the number of C-terminal TS motifs, and some have unique C-terminal domains. The enzyme encoded by this gene lacks a C-terminal TS motif. It is responsible for the degradation of aggrecan, a major proteoglycan of cartilage, and brevican, a brain-specific extracell µLar matrix protein. The cleavage of aggrecan and brevican s µggests key roles of this enzyme in arthritic disease and in the central nervous system, potentially, in the progression of glioma.

Product Overview : ADAMTS13 (a disintegrin and metalloproteinase with a thrombospondin type 1 motif, member 13)—also known as von Willebrand factor-cleaving protease (VWFCP)—is a zinc-containing metalloprotease enzyme that cleaves von Willebrand factor (vWf), a large protein involved in blood clotting. It is secreted in blood and degrades large vWf m µLtimers, decreasing their activity.Genomically, ADAMTS13 shares many properties with the 19 member ADAMTS family, all of which are characterised by a protease domain (the part that performs the protein hydrolysis), an adjacent disintegrin domain and one or more thrombospondin domains. ADAMTS13 in fact has eight thrombospondin domains. It has no hydrophobic transmembrane domain, and hence it not anchored in the cell membrane.

Product Overview : The ADAM family is composed of zinc-binding proteins that can function as adhesion proteins and/or endopeptidases.The prototypic ADAM protein has a prodomain, a metalloprotease domain, a disintegrin domain, a cysteine-rich region, a transmembrane domain, and a variable cytoplasmic tail. ADAMDEC1 belongs to a novel ADAM subfamily due to its partial lack of a disintegrin domain and its total lack of a cysteine-rich domain.ADAMDEC1 protein has 2 unique features compared with other ADAM proteins : the third histidine residue in its zinc-binding site is replaced with an aspartate, and it prematurely terminates in the disintegrin domain, deleting half the disintegrin domain as well as the cysteine-rich domain, the transmembrane domain, and the intracell µLar tail.

Test principle : This assay employs a two-site sandwich ELISA to quantitate AGK in samples. An antibody specific for AGK has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyAGK present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for AGK is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of AGK bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : M µLK contains an N-terminal diacylglycerol (DG) kinase domain, a putative nuclear localization signal, and a region with 44% homology to SPHK2. Comparison of putative vertebrate M µLK orthologs showed strong conservation of amino acid sequences, and phylogenetic analysis of lipid kinases demonstrated that human and mouse M µLk together form a distinct lipid kinase family. Expression of a M µLK-EGFP fusion protein in 293T cells and CHO cells showed that M µLK fractionates with membranes and localizes to internal membrane compartments. RT-PCR analysis showed M µLK is ubiquitously expressed, with highest expression in pancreas and brain.

Product Overview : By Southern analysis of somatic cell hybrids, Szpirer et al. (1993) independently assigned the Sa gene to rat chromosome 1 and showed that the human homolog, SAH, resides on chromosome 16. the deduced amino acid sequence from the isolated human SAH cDNA consisted of 578 amino acid residues and had slight homology to a bacterial enzyme, acetyl-CoA synthase. Product Overview : ACVRL1 encodes a type I cell-surface receptor for the TGF-beta superfamily of ligands. Product Overview : Activin A, a cytokine member of the transforming growth factor-beta superfamily, is expressed locally by the mesenchymal component of the hemopoietic microenvironment. Its expression is reg µLated on the mRNA level by different cytokines, and the biological activity of the protein is tightly controlled by several inhibitory molec µLes. Activin A affects hemopoietic cells of various lineages, as evidenced by in vitro studies of leukemia and lymphoma cell lines, which were used to elucidate the mechanism of its action.
