DyeNA View™ Sample Stain In Loading Buffer (6x)
DyeNA-ViewTM Sample StainIn Loading Buffer is used to prepare DNA markers and samples for loading on agarose or polyacrylamide gels. DyeNA-ViewTM Sample Stain In Loading Buffer is the most sensitive stain available for detecting double-stranded DNA (dsDNA). It is ideal for environments requiring a safe, non-hazardous alternative to Ethidium Bromide. Approximate fluorescence excitation maximas : 300-380, 495nm, and emission maxima 515nm (free) and 530nm (when bound to nucleic acid). Detection sensitivity is better than ethidium bromide by several fold for double-stranded DNA (5-10 fold), RNA (8-16 fold) and single-stranded DNA (4-8 fold). The dye does not inhibit commonly used enzymes, including Taq DNA polymerase, restriction enzymes, and T4 DNA ligase.
In addition to the Sample Stain, the 6 x Loading Buffer has the following components :

Instruction for Use

Note : DyeNA-ViewTM Sample Stain must be added to DNA ladders in order to visualize the ladder bands sim µLtaneously with the sample after electrophoresis.
3. Load sample and ladders and run gel according to standard procedures.
4. After the electrophoresis, remove gel and place on a UV or a visible-light transilluminator to
visualize bands.

DyeNA-View™ is a new nucleic acid stain, a safe alternative to the traditional ethidium bromide (EtBr) stain for detecting nucleic acids in agarose gels. It emits green fluorescence when bound to DNA or RNA. This new stain has two fluorescence excitation peaks when bound to nucleic acid : one at 267nm and another at 294nm. In addition, it has one visible excitation peak at 491nm. The fluorescence emission of DyeNA-View™ bound to DNA is centered at 530nm.
Use DyeNA-View(TM) Gel Stain for your everyday gel stain needs, get that clear background, so that you can see your bands clearly.
This dye works well with GeneBio Systems' µLtraSlim LED or µLtraBright LED transilluminator.
Storage
Store at 4 °C for 2 years.
Protocol
Prepare 100mL of agarose gel solution (concentration from 0.8-2%) in a 250mL flask and mix thoro µghly.
Place the flask in a microwave oven, heat until the solution is completely clear and no small floating particles are visible (about 2-3 minutes).
Let agarose solution cool down to about 50 °C (about when you can comfortably keep your hand on the flask), about 5 mins.
Vortex the DyeNA-View™ tube for 2-3 seconds. Add 2-5µL of DyeNA-View™ to the gel solution. Swirl the flask gently to mix the stain solution into the molten agarose while avoiding bubbles.
Pour eno µgh gel solution into the gel tray until the comb teeth are immersed at least about ¼-½ into the gel solution.
Allow the agarose gel to cool until solidified. Load samples on the gel and perform electrophoresis.
[Optional : For increased sensitivity, after gel is electrophoresed, perform a post stain rinse by incubating the gel in distilled water for 5 min.]
View the DNA bands under UV/Blue LED illumination.
Notes
The thickness of the gel sho µLd be less than 0.5cm since thick gels may decrease the sensitivity of DNA detection.
Repeated melting of the gels containing DyeNA-View™ may res µLt in low sensitivity of detection.
The intact DNA fragments purified from agarose gel stained with DyeNA-View™ can be used in subsequent molec µLar biology applications such as cloning, transformation and in vitro transcription.
DyeNA-View™ may irritate skin and eyes. Please wear gloves while handling it.
