Component of a VCP/p97-AMFR/gp78 complex that participates in the final step of endoplasmic retic µLum-associated degradation (ERAD). The VCP/p97-AMFR/gp78 complex is involved in the sterol-accelerated ERAD degradation of HMGCR thro µgh binding to the HMGCR-INSIG complex at the ER membrane and initiating ubiquitination of HMGCR. The ubiquitinated HMGCR is then released from the ER by the complex into the cytosol for subsequent destruction. Also reg µLates ERAD thro µgh the ubiquitination of UBL4A a component of the BAG6/BAT3 complex. Also acts as a scaffold protein to assemble a complex that couples ubiquitination, retranslocation and deglycosylation. Mediates tumor invasion and metastasis as a receptor for the GPI/autocrine motility factor.

Background : Transcriptional repressor involved in the reg µLation of the circadian rhythm by negatively reg µLating the activity of the clock genes and clock-controlled genes. Acts as the negative limb of a novel autoreg µLatory feedback loop (DEC loop) which differs from the one formed by the PER and CRY transcriptional repressors (PER/CRY loop). Both these loops are interlocked as it represses the expression of PER1 and in turn is repressed by PER1/2 and CRY1/2. Represses the activity of the circadian transcriptional activator : CLOCK-ARNTL/BMAL1 heterodimer by competing for the binding to E-box elements (5\'-CACGTG-3\') found within the promoters of its target genes. Negatively reg µLates its own expression and the expression of DBP and BHLHE41/DEC2. Acts as a corepressor of RXR and the RXR-LXR heterodimers and represses the ligand-induced RXRA/B/G, NR1H3/LXRA, NR1H4 and VDR transactivation activity.
