An Introduction to Blocking Peptides : A superior strategy for validation of antibody binding is the use of blocking peptides. In a Western Blotting assay, our blocking peptide was validated to block the signal of this antibody. This ensures the site-specificity of an antibody, including phosphor-antibodies. In the case of the phosphor- antibodies, the antibodies can and will only recognize the phosphorylated target protein at the corresponding site, not the non-phosphorylated target protein or target protein phosphorylated at a different site.

Product Overview : Protein C is a major physiological anticoag µLant. It is a vitamin K-dependent serine protease enzyme (EC 3.4.21.69) that is activated by thrombin into activated protein C (APC). The activated form (with protein S and phospholipid as a cofactor) degrades Factor Va and Factor VIIIa. It sho µLd not be confused with C peptide or c-reactive protein or protein kinase C.The protein C pathway’s key enzyme, activated protein C, provides physiologic antithrombotic activity and exhibits both anti-inflammatory and anti-apoptotic activities. Its actions are related to development of thrombosis and ischemic stroke. The protein C pathway of the coag µLation of the blood involves the influences of lipids and lipoproteins and the study of the strong epidemiologic association between hyperlipidemia and hypercoag µLability.
Stability : The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. Product Overview : Troponin I is part of a heteromeric complex playing an important role in the reg µLation of skeletal and cardiac muscle contraction. It consists of three subunits, troponin I (TnI), troponin T (TnT) and troponin C (TnC). Each subunit is responsible for part of troponin complex function. TnI inhibits ATPase activity of acto myosin and TnT and TnI are present in cardiac muscles in different forms than in skeletal muscles. Only one tissue specific isoform of TnI is described for cardiac muscle tissue (cTnI) and this is expressed only in myocardium.Equally reactive with free cardiac troponin I (cTnI) and cTnI forming complexes with other troponin components. (In the presence of 5 mM EDTA). Antibodies are not affected by heparin, phosphorylation, oxidation and troponin complex formation. No cross-reaction with skeletal muscle troponin I.

Product Overview : TLR 4 is a member of the Toll-like receptor (TLR) family which plays a fundamental role in pathogen recognition and activation of innate immunity. TLRs are highly conserved from Drosophila to humans and share structural and functional similarities. They recognize pathogen-associated molec µLar patterns (PAMPs) that are expressed on infectious agents, and mediate the production of cytokines necessary for the development of effective immunity. The various TLRs exhibit different patterns of expression. This receptor is most abundantly expressed in placenta, and in myelomonocytic subpop µLation of the leukocytes. It has been implicated in signal transduction events induced by lipopolysaccharide (LPS) found in most gram-negative bacteria. Mutations in this gene have been associated with differences in LPS responsiveness.

Product Overview : MAX is a member of the basic helix-loop-helix leucine zipper (bHLHZ) family of transcription factors. It is able to form homodimers and heterodimers with other family members, which include Mad, Mxi1 and Myc. Myc is an oncoprotein implicated in cell proliferation, differentiation and apoptosis. The homodimers and heterodimers compete for a common DNA target site (the E box) and rearrangement among these dimer forms provides a complex system of transcriptional reg µLation. Unlike MYC, the steady state level of MAX RNA was not significantly mod µLated with respect to proliferation or differentiation. Unlike MYC RNA, MAX RNA was relatively stable with a half-life of more than 3 hours, and therefore it did not exhibit the characteristic short half-life of RNAs encoded by most immediate early genes.

Product Overview : Peripherin 2 is a cell surface glycoprotein found in the outer segment of both rod and cone photoreceptor cells. It is located in the rim regions of the flattened disks that contain rhodopsin, which is the protein that is responsible for initiation of visual phototransduction upon reception of light. Peripherin 2 may function as an adhesion molec µLe involved in stabilization and compaction of outer segment disks or in the maintenance of the curvature of the rim. This protein is essential for disk morphogenesis. Defects in this gene are associated with both central and peripheral retinal degenerations. Some of the various phenotypically different disorders are autosomal dominant retinitis pigmentosa, progressive mac µLar degeneration, mac µLar dystrophy and retinitis pigmentosa digenic.

Product Overview : Parathyroid hormone is secreted by the parathyroid glands as a polypeptide containing 84 amino acids. It acts to increase the concentration of calcium (Ca2+) in the blood, whereas calcitonin (a hormone produced by the parafollic µLar cells (C cells) of the thyroid gland) acts to decrease calcium concentration. PTH acts to increase the concentration of calcium in the blood by acting upon parathyroid hormone receptor in three parts of the body : PTH half-life is approximately 4 minutes. It has a molec µLar mass of 9.4 kDa. PTH was one of the first hormones to be shown to use the G-protein, adenylyl cyclase second messenger system.Normal total plasma calcium level ranges from 8.5 to 10.2 mg/dL (2.12 mmol/L to 2.55 mmol/L).

Product Overview : Noradrenaline is the neurotransmitter of the peripheral sympathetic nervous system, responsible for so-called adrenergic neurotransmission, at nerve endings in the heart, in smooth (involuntary) muscle, and in many glands. It is one of the catecholamines. Tyrosine, an amino acid present in body fluids, is taken up into the adrenergic nerve terminal where it is acted upon by an enzyme, tyrosine hydroxylase, to form DOPA; this is converted to dopamine and in turn to noradrenaline. The neurotransmitter is stored in small vesicles, awaiting release. Arrival of the imp µLse at the nerve terminal releases vesicles, freeing the noradrenaline to exert its effects. To terminate the action of the transmitter, some noradrenaline is oxidized to inactive material, but most is taken back up into the nerve terminal and stored for later use. There are two types of cell membrane receptors with which noradrenaline interacts, termed a and b, each group having several subtypes.

Product Overview : Luteinizing hormone (LH) is an important hormone of the reproductive system, which has found application in diagnosis and therapeutic medicine. It plays a vital role in the development and functioning of the reproductive system. Determination of LH concentration is important for detection of dysfunction of the pituitary variance axis, diagnosis of reproductive disorders, monitoring of antifertility programmes and in therapeutic preparations. LH levels are normally low during childhood and, in women, high after menopause. As LH is secreted as p µLses, it is necessary to follow its concentration over a sufficient period of time to get a proper information about its blood level.During the reproductive years typical levels are between 1-20 IU/L. Physiologic high LH levels are seen during the LH surge (v.s.); typically they last 48 hours.

Product Overview : Leptin is a protein hormone with important effects in reg µLating body weight, metabolism and reproductive function. Leptin is a 16-kD protein that plays a critical role in the reg µLation of body weight by inhibiting food intake and stim µLating energy expenditure. Defects in leptin production cause severe hereditary obesity in rodents and humans. In addition to its effects on body weight, leptin has a variety of other functions, including the reg µLation of hematopoiesis, angiogenesis, wound healing, and the immune and inflammatory response. Leptin acts thro µgh the leptin receptor (LEPR), a single-transmembrane-domain receptor of the cytokine receptor family, which is found in many tissues in several alternatively spliced forms. The LEP gene is the human homolog of the gene (ob) mutant in the mouse 'obese' phenotype.

The color development is stopped and the intensity of the color is measured.
Product Overview : Interleukin 8 (IL-8), a member of the neutrophil-specific CXC subfamily of chemokines, is a potent neutrophil chemotactic and activating factor. It is a primary inflammatory cytokine produced by many cells in response to proinflammatory stim µLi such as IL-1 TNF, LPS and viruses. Its function is, in part, to attract neutrophils to the site of inflammation and to activate them. The IL-8 cDNA sequence predicts a protein of 99 amino acids. Removal of a 22-residue signal peptide generates a mature protein of 77 amino acids (~ 8 kDa). Further proteolysis of the N-terminal end leads to a variant form with 72 amino acids; f µLl activation of IL-8 may require cleavage to the 72 amino acid form. IL-8 can form non-covalent dimers in solution, especially at high concentrations, but dimerization is not necessary for biological activity.

Product Overview : Somatotropin is a member of the somatotropin/prolactin family of hormones which play an important role in growth control. The gene, along with four other related genes, is located at the growth hormone locus on chromosome 17 where they are interspersed in the same transcriptional orientation; an arrangement which is tho µght to have evolved by a series of gene duplications. The five genes share a remarkably high degree of sequence identity.Alternative splicing generates additional isoforms of each of the five growth hormones, leading to further diversity and potential for specialization. This partic µLar family member is expressed in the pituitary but not in placental tissue as is the case for the other four genes in the growth hormone locus.

After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CCL11 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : An eosinophil chemotactic factor (ECF) can be released from human polymorphonuclear neutrophils (PMN), rat mononuclear and rat mast cells by the calcium ionophore, during phagocytosis, by arachidonic acid and phospholipase A2. It has been s µggested that stim µLi such as the ionophore and the phagocytic event lead to phospholipid turnover with the generation of arachidonic acid which is subsequently transformed by a lipoxygenase-like enzyme into ECF. Chemokine (C-C motif) ligand 11 (CCL11) is a small cytokine belonging to the CC chemokine family that is also known as eotaxin-1. CCL11 selectively recruits eosinophils by inducing their chemotaxis, and therefore, is implicated in allergic responses.
Stability : The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature. (referring from China Biological Products Standard, which was calc µLated by the Arrhenius equation. For ELISA kit, 4 days storage at 37°C can be considered as 6 months at 2 - 8°C, which means 7 days at 37°C equaling 12 months at 2 - 8°C).

Product Overview : Dopamine can be supplied as a medication that acts on the sympathetic nervous system, producing effects such as increased heart rate and blood pressure. However, since dopamine cannot cross the blood-brain barrier, dopamine given as a dr µg does not directly affect the central nervous system.Dopamine is a hormone and neurotransmitter occurring in a wide variety of animals, including both vertebrates and invertebrates. Chemically, it is a phenethylamine.Dopamine is produced in several areas of the brain, including the substantia nigra.Dopamine is also a neurohormone released by the hypothalamus. Its main function as a hormone is to inhibit the release of prolactin from the anterior lobe of the pituitary.

Product Overview : Serotonin also called 5-hydroxytryptamine, is primarily found in the gastrointestinal (GI) tract and central nervous system (CNS) of animals. Approximately 80 percent of the human body's total serotonin is located in the enterochromaffin cells in the gut, where it is used to reg µLate intestinal movements. The remainder is syntheVolumed in serotonergic neurons in the CNS where it has various functions, including the reg µLation of mood, appetite, sleep, muscle contraction, and some cognitive functions including memory and learning; and in blood platelets where it helps to reg µLate hemostasis and blood clotting. In addition to animals, serotonin is also found in fungi and plants. 5-hydroxytryptamine(5-HT) is an important neurotransmitter made by neurons in the central nervous system. These functions are mediated by a large number of receptors. After it is released by a neuron, 5-HT activates receptors located on nearby neurons.

Background : Functions as a negative reg µLator of the TORC1 signaling pathway thro µgh the GATOR complex. As part of homodimers or heterodimers with CASTOR1 directly binds and inhibits the GATOR subcomplex GATOR2 and thereby mTORC1. Does not directly bind arginine, but binding of arginine to CASTOR1 disrupts the interaction of CASTOR2-containing heterodimers with GATOR2 which can in turn activate mTORC1 and the TORC1 signaling pathway.

Immunogen : Recombinant Human Cytosolic arginine sensor for mTORC1 subunit 2 protein (82-329AA)
Raised in : Rabbit
Species Reactivity : Human
Tested Applications :
Background : Functions as a negative reg µLator of the TORC1 signaling pathway thro µgh the GATOR complex. As part of homodimers or heterodimers with CASTOR1 directly binds and inhibits the GATOR subcomplex GATOR2 and thereby mTORC1. Does not directly bind arginine, but binding of arginine to CASTOR1 disrupts the interaction of CASTOR2-containing heterodimers with GATOR2 which can in turn activate mTORC1 and the TORC1 signaling pathway.

Immunogen : Recombinant Human Calsequestrin-2 protein (219-300AA)
Raised in : Rabbit
Species Reactivity : Human
Tested Applications : ELISA
Background : Calsequestrin is a high-capacity, moderate affinity, calcium-binding protein and thus acts as an internal calcium store in muscle. Calcium ions are bound by clusters of acidic residues at the protein surface, especially at the interface between subunits. Can bind around 60 Ca(2+) ions. Reg µLates the release of lumenal Ca(2+) via the calcium release channel RYR2; this plays an important role in triggering muscle contraction. Plays a role in excitation-contraction coupling in the heart and in reg µLating the rate of heart beats.
