Background : This gene is a proto-oncogene that encodes a RING finger E3 ubiquitin ligase. The encoded protein is one of the enzymes required for targeting substrates for degradation by the proteasome. This protein mediates the transfer of ubiquitin from ubiquitin conj µgating enzymes (E2) to specific substrates. This protein also contains an N-terminal phosphotyrosine binding domain that allows it to interact with numerous tyrosine-phosphorylated substrates and target them for proteasome degradation. As such it functions as a negative reg µLator of many signal transduction pathways. This gene has been found to be mutated or translocated in many cancers including acute myeloid leukaemia, and expansion of CGG repeats in the 5' UTR has been associated with Jacobsen syndrome. Background : The scaffolding protein encoded by Caveolin-1 is the main component of the caveolae plasma membranes found in most cell types. The protein links integrin subunits to the tyrosine kinase FYN, an initiating step in coupling integrins to the Ras-ERK pathway and promoting cell cycle progression. An Introduction to Blocking Peptides : A superior strategy for validation of antibody binding is the use of blocking peptides. In a Western Blotting assay, our blocking peptide was validated to block the signal of this antibody. This ensures the site-specificity of an antibody, including phosphor-antibodies. In the case of the phosphor- antibodies, the antibodies can and will only recognize the phosphorylated target protein at the corresponding site, not the non-phosphorylated target protein or target protein phosphorylated at a different site.
