Product Overview : High molec µLar weight kininogen (HMWK) plays an important role in assembly of the plasma kallikrein -kinin system. The KNG1 gene generates both HMWK and low molec µLar weight kininogen (LMWK) thro µgh alternative splicing. Both HMWK and LMWK contain an identical heavy chain consisting of protein domains 1 2, and 3. However, HMWK contains a 56-kD light chain that consists of domains 5 and 6H, whereas LMWK contains a unique 4-kD light chain that consists of domain 5L. In both proteins, the heavy and light chains are linked by domain 4, which contains the bradykinin (BK) nonapeptide. BK, which is released by plasma kallikrein, is a potent inflammatory mediator that causes vasodilation and enhanced capillary permeability, induces pain, and stim µLates production of nitric oxide and prostacyclin from endothelial cells.

Product Overview : The RAB6KIFL gene spans approximately 8.5 kb of genomic DNA and contains 19 exons. The composite cDNA encodes a deduced 890-amino acid protein that contains a motor domain and an ATP catalytic domain, hallmarks of kinesin-like proteins, in the N terminus. The protein shares 87% sequence identity with the mouse Rab6kifl protein. In contrast to the ubiquitous expression of the mouse protein, expression of the human, approximately 3.4-kb RAB6KIFL transcript appears to be tissue specific. Northern blot analysis detected high expression in fetal tissues and in ad µLt thymus, bone marrow, and testis, and low expression in heart, placenta, and spleen. Product Overview : Using primers known to amplify HLA-C-specific NK receptors to screen clones with NK triggering activity, Bottino et al. (1996) also isolated a KIR2DS4 cDNA, which they termed KKA3. Sequence analysis revealed that KIR2DS4 encodes a deduced 304-amino acid protein with an extracell µLar domain similar to that of KIR2D p58 receptors; however, in the transmembrane region, there is a charged lysine residue, and the cytoplasmic tail has only 39 amino acids with no ITIM (immunoreceptor tyrosine-based inhibitory motif). KIRs with short cytoplasmic tails are associated with NK cell triggering rather than inhibition. Immunoprecipitation analysis showed that KIR2DS4 is expressed in 2 forms, one of 35 to 45 kD and the other of 55 to 58 kD. Flow cytometry analysis showed that KIR2DS4, unlike KIR2DS1 and KIR2DS2, was expressed in only a minority of individuals tested.

Product Overview : KRT33A is a member of the keratin gene family. It is one of the type I hair keratin genes which are clustered in a region of chromosome 17q12-q21 and have the same direction of transcription. As a type I hair keratin, it is an acidic protein which heterodimerizes with type II keratins to form hair and nails. There are two isoforms of this protein, encoded by two separate genes, KRTHA3A and KRTHA3B. Rogers et al. (1998) reported that the deduced KRTHA3A protein has 405 amino acids and shares 93.3% amino acid sequence identity with KRTHA3B. By RT-PCR, Rogers et al. Test principle : This assay employs a two-site sandwich ELISA to quantitate KCNQ1DN in samples. An antibody specific for KCNQ1DN has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyKCNQ1DN present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for KCNQ1DN is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of KCNQ1DN bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : Imprinting is a phenomenon in which epigenetic modifications lead to expression or suppression of alleles of some genes based on their parental origin. Wilms tumor-2 (WT2) is defined by maternal-specific loss of heterozygosity of a critical region on chromosome 11p15.5 that includes several imprinted genes. KCNQ1DN is an imprinted gene located within the WT2 critical region that is expressed from the maternal allele. KCNQ1DN has a small ORF encoding 68 amino acids, but it lacks a Kozak consensus sequence around the initiator ATG, s µggesting it is not translated. KCNQ1DN expression was present in fetal kidney between 82 and 103 days of gestation. RT-PCR showed monoallelic expression of KCNQ1DN in fetal kidney and maternal expression of KCNQ1DN in placenta.
