Test principle : This assay employs a two-site sandwich ELISA to quantitate CYP2C19 in samples. An antibody specific for CYP2C19 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyCYP2C19 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for CYP2C19 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CYP2C19 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : CYP2C19 is an enzyme. This protein, a member of the cytochrome P450 mixed-function oxidase system, is involved in the metabolism of xenobiotics, including many proton pump inhibitors and antiepileptics. CYP2C19 is a liver enzyme that acts on 10-15% of dr µgs in current clinical use, including the antiplatelet clopidogrel, dr µgs that treat pain associated with µLcers, such as omeprazole, antiseizure dr µgs such as mephenytoin, the antimalarial proguanil, and the anxiolytic diazepam. The gene encodes a member of the cytochrome P450 superfamily of enzymes. These proteins are monooxygenases that catalyze many reactions involved in dr µg metabolism and synthesis of cholesterol, steroids and other lipids. This protein localizes to the endoplasmic retic µLum and is known to metabolize many dr µgs.
Stability : The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature.
