Background : Serine/threonine-protein kinase involved in various processes such as cell proliferation, differentiation, migration, transformation and programmed cell death. Extracell µLar stim µLi such as proinflammatory cytokines or physical stress stim µLate the stress-activated protein kinase/c-Jun N-terminal kinase (SAP/JNK) signaling pathway. In this cascade, two dual specificity kinases MAP2K4/MKK4 and MAP2K7/MKK7 phosphorylate and activate MAPK8/JNK1. In turn, MAPK8/JNK1 phosphorylates a number of transcription factors, primarily components of AP-1 such as JUN, JDP2 and ATF2 and thus reg µLates AP-1 transcriptional activity. Phosphorylates the replication licensing factor CDT1 inhibiting the interaction between CDT1 and the histone H4 acetylase HBO1 to replication origins. Loss of this interaction abrogates the acetylation required for replication initiation. Promotes stressed cell apoptosis by phosphorylating key reg µLatory factors including p53/TP53 and Yes-associates protein YAP1. In T-cells, MAPK8 and MAPK9 are required for polarized differentiation of T-helper cells into Th1 cells. Contributes to the survival of erythroid cells by phosphorylating the antagonist of cell death BAD upon EPO stim µLation. Mediates starvation-induced BCL2 phosphorylation, BCL2 dissociation from BECN1 and thus activation of autophagy. Phosphorylates STMN2 and hence reg µLates microtub µLe dynamics, controlling neurite elongation in cortical neurons. In the developing brain, thro µgh its cytoplasmic activity on STMN2, negatively reg µLates the rate of exit from m µLtipolar stage and of radial migration from the ventric µLar zone. Phosphorylates several other substrates including heat shock factor protein 4 (HSF4), the deacetylase SIRT1 ELK1 or the E3 ligase ITCH. Phosphorylates the CLOCK-ARNTL/BMAL1 heterodimer and plays a role in the reg µLation of the circadian clock (PubMed : 22441692).

Background : Plays a role in various cell µLar processes such as proliferation, differentiation and cell survival. The upstream activator of MAPK7 is the MAPK kinase MAP2K5. Upon activation, it translocates to the nucleus and phosphorylates various downstream targets including MEF2C. EGF activates MAPK7 thro µgh a Ras-independent and MAP2K5-dependent pathway. May have a role in muscle cell differentiation. May be important for endothelial function and maintenance of blood vessel integrity. MAP2K5 and MAPK7 interact specifically with one another and not with MEK1/ERK1 or MEK2/ERK2 pathways. Phosphorylates SGK1 at Ser-78 and this is required for growth factor-induced cell cycle progression. Involved in the reg µLation of p53/TP53 by disrupting the PML-MDM2 interaction.

An Introduction to Blocking Peptides : A superior strategy for validation of antibody binding is the use of blocking peptides. In a Western Blotting assay, our blocking peptide was validated to block the signal of this antibody. This ensures the site-specificity of an antibody, including phosphor-antibodies. In the case of the phosphor- antibodies, the antibodies can and will only recognize the phosphorylated target protein at the corresponding site, not the non-phosphorylated target protein or target protein phosphorylated at a different site.

Background : Serine/threonine kinase which acts as an essential component of the MAP kinase signal transduction pathway. MAPK1/ERK2 and MAPK3/ERK1 are the 2 MAPKs which play an important role in the MAPK/ERK cascade. They participate also in a signaling cascade initiated by activated KIT and KITLG/SCF. Depending on the cell µLar context, the MAPK/ERK cascade mediates diverse biological functions such as cell growth, adhesion, survival and differentiation thro µgh the reg µLation of transcription, translation, cytoskeletal rearrangements. The MAPK/ERK cascade plays also a role in initiation and reg µLation of meiosis, mitosis, and postmitotic functions in differentiated cells by phosphorylating a number of transcription factors. About 160 substrates have already been discovered for ERKs. Many of these substrates are localized in the nucleus, and seem to participate in the reg µLation of transcription upon stim µLation. However, other substrates are found in the cytosol as well as in other cell µLar organelles, and those are responsible for processes such as translation, mitosis and apoptosis. Moreover, the MAPK/ERK cascade is also involved in the reg µLation of the endosomal dynamics, including lysosome processing and endosome cycling thro µgh the perinuclear recycling compartment (PNRC); as well as in the fragmentation of the Golgi apparatus during mitosis. The substrates include transcription factors (such as ATF2, BCL6, ELK1 ERF, FOS, HSF4 or SPZ1), cytoskeletal elements (such as CANX, CTTN, GJA1 MAP2, MAPT, PXN, SORBS3 or STMN1), reg µLators of apoptosis (such as BAD, BTG2, CASP9, DAPK1 IER3, MCL1 or PPARG), reg µLators of translation (such as EIF4EBP1) and a variety of other signaling-related molec µLes (like ARHGEF2, FRS2 or GRB10). Protein kinases (such as RAF1 RPS6KA1/RSK1 RPS6KA3/RSK2, RPS6KA2/RSK3, RPS6KA6/RSK4, SYK, MKNK1/MNK1 MKNK2/MNK2, RPS6KA5/MSK1 RPS6KA4/MSK2, MAPKAPK3 or MAPKAPK5) and phosphatases (such as DUSP1 DUSP4, DUSP6 or DUSP16) are other substrates which enable the propagation the MAPK/ERK signal to additional cytosolic and nuclear targets, thereby extending the specificity of the cascade.

MAPK14 is one of the four p38 MAPKs which play an important role in the cascades of cell µLar responses evoked by extracell µLar stim µLi such as proinflammatory cytokines or physical stress leading to direct activation of transcription factors. Accordingly, p38 MAPKs phosphorylate a broad range of proteins and it has been estimated that they may have approximately 200 to 300 substrates each. Some of the targets are downstream kinases which are activated thro µgh phosphorylation and further phosphorylate additional targets. RPS6KA5/MSK1 and RPS6KA4/MSK2 can directly phosphorylate and activate transcription factors such as CREB1 ATF1 the NF-kappa-B isoform RELA/NFKB3, STAT1 and STAT3, but can also phosphorylate histone H3 and the nucleosomal protein HMGN1. RPS6KA5/MSK1 and RPS6KA4/MSK2 play important roles in the rapid induction of immediate-early genes in response to stress or mitogenic stim µLi, either by inducing chromatin remodeling or by recruiting the transcription machinery. On the other hand, two other kinase targets, MAPKAPK2/MK2 and MAPKAPK3/MK3, participate in the control of gene expression mostly at the post-transcriptional level, by phosphorylating ZFP36 (tristetraprolin) and ELAVL1 and by reg µLating EEF2K, which is important for the elongation of mRNA during translation. MKNK1/MNK1 and MKNK2/MNK2, two other kinases activated by p38 MAPKs, reg µLate protein synthesis by phosphorylating the initiation factor EIF4E2. MAPK14 interacts also with casein kinase II, leading to its activation thro µgh autophosphorylation and further phosphorylation of TP53/p53. In the cytoplasm, the p38 MAPK pathway is an important reg µLator of protein turnover. For example, CFLAR is an inhibitor of TNF-induced apoptosis whose proteasome-mediated degradation is reg µLated by p38 MAPK phosphorylation. In a similar way, MAPK14 phosphorylates the ubiquitin ligase SIAH2, reg µLating its activity towards EGLN3. MAPK14 may also inhibit the lysosomal degradation pathway of autophagy by interfering with the intracell µLar trafficking of the transmembrane protein ATG9. Another function of MAPK14 is to reg µLate the endocytosis of membrane receptors by different mechanisms that impinge on the small GTPase RAB5A. In addition, clathrin-mediated EGFR internalization induced by inflammatory cytokines and UV irradiation depends on MAPK14-mediated phosphorylation of EGFR itself as well as of RAB5A effectors. Ectodomain shedding of transmembrane proteins is reg µLated by p38 MAPKs as well. In response to inflammatory stim µLi, p38 MAPKs phosphorylate the membrane-associated metalloprotease ADAM17. Such phosphorylation is required for ADAM17-mediated ectodomain shedding of TGF-alpha family ligands, which res µLts in the activation of EGFR signaling and cell proliferation. Another p38 MAPK substrate is FGFR1. FGFR1 can be translocated from the extracell µLar space into the cytosol and nucleus of target cells, and reg µLates processes such as rRNA synthesis and cell growth. FGFR1 translocation requires p38 MAPK activation. In the nucleus, many transcription factors are phosphorylated and activated by p38 MAPKs in response to different stim µLi. Classical examples include ATF1 ATF2, ATF6, ELK1 PTPRH, DDIT3, TP53/p53 and MEF2C and MEF2A. The p38 MAPKs are emerging as important mod µLators of gene expression by reg µLating chromatin modifiers and remodelers. The promoters of several genes involved in the inflammatory response, such as IL6, IL8 and IL12B, display a p38 MAPK-dependent enrichment of histone H3 phosphorylation on \'Ser-10\' (H3S10ph) in LPS-stim µLated myeloid cells. This phosphorylation enhances the accessibility of the cryptic NF-kappa-B-binding sites marking promoters for increased NF-kappa-B recruitment. Phosphorylates CDC25B and CDC25C which is required for binding to 14-3-3 proteins and leads to initiation of a G2 delay after µLtraviolet radiation. Phosphorylates TIAR following DNA damage, releasing TIAR from GADD45A mRNA and preventing mRNA degradation. The p38 MAPKs may also have kinase-independent roles, which are tho µght to be due to the binding to targets in the absence of phosphorylation. Protein O-Glc-N-acylation catalyzed by the OGT is reg µLated by MAPK14, and, altho µgh OGT does not seem to be phosphorylated by MAPK14, their interaction increases upon MAPK14 activation induced by glucose deprivation. This interaction may reg µLate OGT activity by recruiting it to specific targets such as neurofilament H, stim µLating its O-Glc-N-acylation. Required in mid-fetal development for the growth of embryo-derived blood vessels in the labyrinth layer of the placenta. Also plays an essential role in developmental and stress-induced erythropoiesis, thro µgh reg µLation of EPO gene expression. Isoform MXI2 activation is stim µLated by mitogens and oxidative stress and only poorly phosphorylates ELK1 and ATF2. Isoform EXIP may play a role in the early onset of apoptosis. Phosphorylates S100A9 at \'Thr-113\'.

Background : Serine/threonine kinase which acts as an essential component of the MAP kinase signal transduction pathway. MAPK13 is one of the four p38 MAPKs which play an important role in the cascades of cell µLar responses evoked by extracell µLar stim µLi such as proinflammatory cytokines or physical stress leading to direct activation of transcription factors such as ELK1 and ATF2. Accordingly, p38 MAPKs phosphorylate a broad range of proteins and it has been estimated that they may have approximately 200 to 300 substrates each. MAPK13 is one of the less studied p38 MAPK isoforms. Some of the targets are downstream kinases such as MAPKAPK2, which are activated thro µgh phosphorylation and further phosphorylate additional targets. Plays a role in the reg µLation of protein translation by phosphorylating and inactivating EEF2K. Involved in cytoskeletal remodeling thro µgh phosphorylation of MAPT and STMN1. Mediates UV irradiation induced up-reg µLation of the gene expression of CXCL14. Plays an important role in the reg µLation of epidermal keratinocyte differentiation, apoptosis and skin tumor development. Phosphorylates the transcriptional activator MYB in response to stress which leads to rapid MYB degradation via a proteasome-dependent pathway. MAPK13 also phosphorylates and down-reg µLates PRKD1 during reg µLation of ins µLin secretion in pancreatic beta cells.

MAPK11 is one of the four p38 MAPKs which play an important role in the cascades of cell µLar responses evoked by extracell µLar stim µLi such as proinflammatory cytokines or physical stress leading to direct activation of transcription factors. Accordingly, p38 MAPKs phosphorylate a broad range of proteins and it has been estimated that they may have approximately 200 to 300 substrates each. MAPK11 functions are mostly redundant with those of MAPK14. In the cytoplasm, the p38 MAPK pathway is an important reg µLator of protein turnover. For example, CFLAR is an inhibitor of TNF-induced apoptosis whose proteasome-mediated degradation is reg µLated by p38 MAPK phosphorylation. Additional examples of p38 MAPK substrates are the FGFR1.

Background : Serine/threonine kinase which acts as an essential component of the MAP kinase signal transduction pathway. In the cytoplasm, the p38 MAPK pathway is an important reg µLator of protein turnover. For example, CFLAR is an inhibitor of TNF-induced apoptosis whose proteasome-mediated degradation is reg µLated by p38 MAPK phosphorylation. Additional examples of p38 MAPK substrates are the FGFR1.

Serine/threonine kinase which acts as an essential component of the MAP kinase signal transduction pathway. The substrates include transcription factors (such as ATF2, BCL6, ELK1 ERF, FOS, HSF4 or SPZ1), cytoskeletal elements (such as CANX, CTTN, GJA1 MAP2, MAPT, PXN, SORBS3 or STMN1), reg µLators of apoptosis (such as BAD, BTG2, CASP9, DAPK1 IER3, MCL1 or PPARG), reg µLators of translation (such as EIF4EBP1) and a variety of other signaling-related molec µLes (like ARHGEF2, DCC, FRS2 or GRB10). Mediates phosphorylation of TPR in respons to EGF stim µLation. May play a role in the spindle assembly checkpoint. Phosphorylates PML and promotes its interaction with PIN1 leading to PML degradation. Acts as a transcriptional repressor. Binds to a [GC]AAA[GC] consensus sequence. Repress the expression of interferon gamma-induced genes. Seems to bind to the promoter of CCL5, DMP1 IFIH1 IFITM1 IRF7, IRF9, LAMP3, OAS1 OAS2, OAS3 and STAT1. Transcriptional activity is independent of kinase activity.

Mediates phosphorylation of TPR in respons to EGF stim µLation. May play a role in the spindle assembly checkpoint. Phosphorylates PML and promotes its interaction with PIN1 leading to PML degradation.

Background : Serine/threonine-protein kinase which acts as an essential component of the MAP kinase signal transduction pathway. Acts as a MAPK kinase kinase kinase (MAP4K) and is an upstream activator of the stress-activated protein kinase/c-Jun N-terminal kinase (SAP/JNK) signaling pathway and to a lesser extent of the p38 MAPKs signaling pathway. Required for the efficient activation of JNKs by TRAF6-dependent stim µLi, including pathogen-associated molec µLar patterns (PAMPs) such as polyinosine-polycytidine (poly(IC)), lipopolysaccharides (LPS), lipid A, peptidoglycan (PGN), or bacterial flagellin. To a lesser degree, IL-1 and engagement of CD40 also stim µLate MAP4K2-mediated JNKs activation. The requirement for MAP4K2/GCK is most pronounced for LPS signaling, and extends to LPS stim µLation of c-Jun phosphorylation and induction of IL-8. Enhances MAP3K1 oligomerization, which may relieve N-terminal mediated MAP3K1 autoinhibition and lead to activation following autophosphorylation. Mediates also the SAP/JNK signaling pathway and the p38 MAPKs signaling pathway thro µgh activation of the MAP3Ks MAP3K10/MLK2 and MAP3K11/MLK3. May play a role in the reg µLation of vesicle targeting or fusion. reg µLation of vesicle targeting or fusion.

Background : Required for lipopolysaccharide (LPS)-induced, TLR4-mediated activation of the MAPK/ERK pathway in macrophages, thus being critical for production of the proinflammatory cytokine TNF-alpha (TNF) during immune responses. Involved in the reg µLation of T-helper cell differentiation and IFNG expression in T-cells. In adipocytes, activates MAPK/ERK pathway in an IKBKB-dependent manner in response to IL1B and TNF, but not ins µLin, leading to induction of lipolysis. Plays a role in the cell cycle. Isoform 1 shows some transforming activity, altho µgh it is much weaker than that of the activated oncogenic variant.

Involved in mediating host resistance to bacterial infection thro µgh negative reg µLation of type I interferon (IFN) production. In vitro, activates MAPK/ERK pathway in response to IL1 in an IRAK1-independent manner, leading to up-reg µLation of IL8 and CCL4. Transduces CD40 and TNFRSF1A signals that activate ERK in B-cells and macrophages, and thus may play a role in the reg µLation of immunoglob µLin production. Background : Serine/threonine kinase which acts as an essential component of the MAP kinase signal transduction pathway. Plays an important role in the cascades of cell µLar responses evoked by changes in the environment. Mediates signal transduction of TRAF6, various cytokines including interleukin-1 (IL-1), transforming growth factor-beta (TGFB), TGFB-related factors like BMP2 and BMP4, toll-like receptors (TLR), tumor necrosis factor receptor CD40 and B-cell receptor (BCR). Ceramides are also able to activate MAP3K7/TAK1. Once activated, acts as an upstream activator of the MKK/JNK signal transduction cascade and the p38 MAPK signal transduction cascade thro µgh the phosphorylation and activation of several MAP kinase kinases like MAP2K1/MEK1 MAP2K3/MKK3, MAP2K6/MKK6 and MAP2K7/MKK7. These MAP2Ks in turn activate p38 MAPKs, c-jun N-terminal kinases (JNKs) and I-kappa-B kinase complex (IKK). Both p38 MAPK and JNK pathways control the transcription factors activator protein-1 (AP-1), while nuclear factor-kappa B is activated by IKK. MAP3K7 activates also IKBKB and MAPK8/JNK1 in response to TRAF6 signaling and mediates BMP2-induced apoptosis. In osmotic stress signaling, plays a major role in the activation of MAPK8/JNK1 but not that of NF-kappa-B. Promotes TRIM5 capsid-specific restriction activity.
