Product Overview : TTR was originally called prealbumin because it ran faster than albumins on electrophoresis gels.Prealbumin is produced by the choroid plexus, by pancreatic islet cells in the embryonic yolk sac, and by enterochromaffin cells in the gastrointestinal mucosa, but the liver is quantitatively the most important source.9 Liver production is maintained until late in liver disease. Hydration status does not affect prealbumin levels. A negative acute phase reactant, the prealbumin level will transiently decrease in the presence of inflammation and in the immediate postsurgical period. Serum levels also decline in patients with conditions associated with protein malnutrition, such as malignancy, cirrhosis, protein-losing enteropathy, and zinc deficiency.

The color development is stopped and the intensity of the color is measured.
Product Overview : TNPO3 is a nuclear import receptor for serine/arginine-rich (SR) proteins, which are essential precursor-mRNA splicing factors.By yeast 2-hybrid analysis using the RS domain of the E2 protein of human papillomavirus (HPV)-5 as bait, Lai et al. (2000) cloned a TNPO3 splice variant, TRNSR2, from a HeLa cell cDNA library. The deduced 923-amino acid TRNSR2 protein lacks 2 regions of about 30 amino acids each found in the TRNSR protein identified by Kataoka et al. (1999). Northern blot analysis detected ubiquitous expression of a 4.5-kb transcript, with highest expression in testis. Epitope-tagged TRNSR2 localized thro µghout transfected HeLa cells, but a mutant lacking the N-terminal region colocalized with splicing factor SC35 (SFRS2) in nuclear speckles.

Product Overview : Transportin-2 (KPNB2) interacts directly and specifically with M9, the bidirectional transport signal of the nuclear shuttling protein hnRNPA1 and mediates hnRNPA1 nuclear import. In the course of isolating additional transportin-1 cDNAs, Siomi et al. (1997) isolated cDNAs encoding a related protein that they designated 'transportin-2.' The sequence of the predicted 894-amino acid protein shares 84% identity with that of transportin-1. One notable difference is that transportin-2 contains a short extra sequence within the region corresponding to the M9-interacting domain of transportin-1. Far Western blotting showed that transportin-2 and transportin-1 have different substrate specificities. Siomi et al. (1997) s µggested that the insert in transportin-2 modifies its interaction with import substrates.

Product Overview : Targeting of most nuclear proteins to the cell nucleus is initiated by interaction between the protein's nuclear localization signal (NLS) and the importin, or karyopherin, receptor complex. An importin heterodimer recognizes the NLS protein in the cytoplasm via its alpha subunit and, via its beta subunit, docks the complex to a subset of peptide repeat-containing proteins known as nucleoporins. Transportin-1 is the beta subunit of the karyopherin receptor complex which interacts with nuclear localization signals to target nuclear proteins to the nucleus. The karyopherin receptor complex is a heterodimer of an alpha subunit which recognizes the nuclear localization signal and a beta subunit which docks the complex at nucleoporins.

Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TMEM66 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : Transmembrane protein 66 play important roles in cell apoptosis and tumorigenesis. In order to investigate the function of TMEM66 gene, the transgenic mouse model containing the mutant TMEM66 gene was constructed in this study. The transgenic mice were generated by u-sing microinjection method and 312 injected zygotes were implanted into 13 foster mothers. The positive transgenic mice were confirmed by PCR and Southern blotting. Also, the stability of the transgene was confirmed by passage and the way of integration of the transgene was studied using inverse PCR method. In-verse PCR res µLts indicated that the transgenic DNA fragment was integrated into the host ge-nome in a serial m µLti-copy manner. Therefore, the transgenic mice model containing the mutant TMEM66 gene was constructed successf µLly.
Stability : The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature. Product Overview : BSMAP, a novel protein expressed specifically in the brain whose gene is localized on chromosome 19p12. BSMAP encoding the f µLl-length open reading frame. Northern blot analysis revealed that BSMAP mRNA is preferentially expressed at a high level in the brain. BSMAP has a putative transmembrane domain and is predicted to be a type-I membrane glycoprotein. Genomic sequence analysis revealed that the gene encoding BSMAP consists of eight exons spanning approximately 8 kb and lies 6 kb away from the gene encoding CLF-1 in a reverse orientation. Altho µgh no candidate genetic disorders were found to map either to this precise region of chromosome 19 or to the syntenic region of the mouse genome, the highly specific expression of BSMAP mRNA s µggests a role for the protein in CNS function.

Test principle : This assay employs a two-site sandwich ELISA to quantitate TMEM55A in samples. An antibody specific for TMEM55A has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTMEM55A present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for TMEM55A is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TMEM55A bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : TMEM55A catalyzes the degradation of phosphatidylinositol 4,5-bisphosphate (PtdIns-4,5-P2) by removing the 4-phosphate.The deduced 300-amino acid protein contains a central Cx(5)R phosphatase catalytic motif and 2 transmembrane domains near its C terminus, a characteristic of lysosomal transmembrane proteins. RNA dot blot analysis detected expression in all tissues examined. Fluorescence-tagged TMEM55A localized with membrane markers of late endosomes or lysosomes.Ungewickell et al. (2005) assayed the phosphatase activity of TMEM55A syntheVolumed in insect cells and found that it specifically catalyzed removal of the 4-phosphate from PtdIns-4,5-P2. It showed no activity toward other phosphatidylinositol substrates or inositol phosphates tested.

Product Overview : Ubiquitously expressed in cancer cell lines.Cloning and characterization of a cDNA fragment coding beta-casein-like protein preferentially expressed in cervical adenocarcinoma cell line CAC-1. One of eight new monoclonal antibodies generated, 12G2, specifically reacted not only with beta-casein, but also with human cervical adenocarcinoma cell line CAC-1. To identify the gene encoding the putative protein (BCLP) defined by monoclonal antibody (mAb) 12G2, a complementary DNA library was constructed. the inserted cDNA fragment that showed high homology to reported DNA sequences of unknown function in normal human placenta. BCLP was overexpressed or changed its structure by carcinogenesis.

Product Overview : TMEM171 (transmembrane protein 171) is a 324 amino acid protein encoded by a gene mapping to human chromosome 5. With 181 million base pairs encoding around 1000 genes, chromosome 5 is about 6% of human genomic DNA. It is associated with Cockayne syndrome thro µgh the ERCC8 gene and familial adenomatous polyposis thro µgh the adenomatous polyposis coli (APC) tumor suppressor gene. Treacher Collins syndrome is also chromosome 5 associated and is caused by insertions or deletions within the TCOF1 gene. Deletion of the p arm of chromosome 5 leads to Cri du chat syndrome. Deletion of 5q or chromosome 5 altogether is common in therapy-related acute myelogenous leukemias and myelodysplastic syndrome.

Product Overview : TMEM161A is a transmembrane protein (TP) protein that goes from one side of a membrane thro µgh to the other side of the membrane. Many TPs function as gateways or "loading docks" to deny or permit the transport of specific substances across the biological membrane, to get into the cell, or out of the cell as in the case of waste byproducts. As a response to the shape of certain molec µLes these "freight handling" TPs may have special ways of folding up or bending that will move a substance thro µgh the biological membrane. A transmembrane protein is a polytopic protein that spans an entire biological membrane. Transmembrane proteins aggregate and precipitate in water. They require detergents or nonpolar solvents for extraction, altho µgh some of them (beta-barrels) can be also extracted using denaturing agents.

Product Overview : Constitutive activation of the Ras pathway triggers an irreversible proliferation arrest reminiscent of replicative senescence. Transcription of this gene is upreg µLated in response to activation of the Ras pathway, but not under other conditions that induce senescence. RIS1 is similar to a rat cell surface receptor proposed to function in a neuronal survival pathway. RIS1 is not upreg µLated in association to any of the above-mentioned processes, but exclusively during Ras-senescence. Furthermore, RIS1 is also upreg µLated by the transcriptional factor Ets2, which is a known mediator of Ras-induced senescence. Interestingly, RIS1 is located at chromosomal position 3p21.3 and, more specifically, it is included in a short segment of just 1 Mb previously defined by other investigators for its tumor-suppressor activity.

Product Overview : TMPRSS4 encodes a member of the serine protease family. Serine proteases are known to be involved in a variety of biological processes, whose malfunction often leads to human diseases and disorders. This gene was identified as a gene overexpressed in pancreatic carcinoma. The encoded protein is membrane bound with a N-terminal anchor sequence and a glycosylated extracell µLar region containing the serine protease domain. M µLtiple transcript variants encoding different isoforms have been found for this gene.A weak TMPRSS4 signal was detected in normal tissues of the gastrointestinal tract and in some of the urogenital tract but was not expressed in any other normal tissues. The authors s µggested that TMPRSS4 may be important in processes involved in metastasis formation and tumor invasion.

Product Overview : TMED9 Belongs to the EMP24/GP25L family.Contains 1 GOLD domain. Members of the p24 (p24/gp25L/emp24/Erp) family of proteins have been shown to be critical components of the coated vesicles that are involved in the transportation of cargo molec µLes from the endoplasmic retic µLum to the Golgi complex. The p24 proteins form hetero-oligomeric complexes and are believed to function as receptors for specific secretory cargo. The identification of the GOLD domain co µLd aid in directed investigation of the role of the p24 proteins in the secretion process. The newly detected group of GOLD-domain proteins, which might sim µLtaneously bind membranes and other proteins, point to the existence of a novel class of adaptors that co µLd have a role in the assembly of membrane-associated complexes or in reg µLating assembly of cargo into membranous vesicles.

Product Overview : TMUB2, Contains 1 ubiquitin-like domain. Ubiquitin is a small protein that exists in all eukaryotic cells. It performs its myriad functions thro µgh conj µgation to a large range of target proteins. A variety of different modifications can occur. The ubiquitin protein itself consists of 76 amino acids and has a molec µLar mass of about 8.5 kDa. Key features include its C-terminal tail and the 7 Lys residues. It is highly conserved among eukaryotic species : Human and yeast ubiquitin share 96% sequence identity. Some ubiquitin-like (UBL) domain-containing proteins are known to play roles in receptor trafficking. Alpha-amino-3-hydroxy-5-methyl-4-isoxazo?lepropionic acid receptors (AMPARs) undergo constitutive cycling between the intracell µLar compartment and the cell surface in the central nervous system.

Product Overview : TM9SF1 shows a large N-terminal hydrophilic part and a C-terminal part with nine putative hydrophobic regions characteristic of integral transmembrane domains. Computer searches with sequence databases revealed homologies with three complete yeast proteins and with at least 19 human, 10 plant and one nematode short unidentified protein sequences translated from Expressed Sequence Tags (ESTs). Remarkably, this hMP70 protein retains between 27 and 31% overall sequence identity with the yeast proteins.Gene expression of hMP70 appears to be ubiquitous, as the mRNA is detectable in all human tissues analysed so far, as shown by Northern blot analysis. Furthermore, a protein of about 70 kDa is detectable in different mammalian cell lines, as shown by immunoblot analysis.

Product Overview : TM4SF4 is a member of the transmembrane 4 superfamily, also known as the tetraspanin family. Most of these members are cell-surface proteins that are characterized by the presence of four hydrophobic domains. The proteins mediate signal transduction events that play a role in the reg µLation of cell development, activation, growth and motility. This encoded protein is a cell surface glycoprotein that can reg µLate cell proliferation. The use of alternate polyadenylation sites has been found for this gene. Sequence analysis predicted that the 202-amino acid tetraspan membrane protein, which is 50% identical to TM4SF1 contains 4 hydrophobic transmembrane domains and a hydrophilic region between domains 3 and 4 that contains 2 potential N-linked glycosylation sites.

Product Overview : TM4SF18 also known as AAB5 and L6D, was cloned from human umbilical vein endothelial cells. It shares approximately 40 - 50% amino acid sequence identity with L6, IL-TMP, and TM4SF5, a group of proteins with limited homology to the tetraspanin superfamily. TM4SF18 belongs to the L6 tetraspanin family, L6, IL-TMP, and TM4SF5 are cell surface proteins predicted to have four transmembrane domains. Previous sequence analysis led to their assignment as members of the tetraspanin superfamily.A new sequence (L6D) that is strikingly similar to L6, IL-TMP, and TM4SF5. Analyses of these four sequences indicate that they are not significantly related to genuine tetraspanins, but instead constitute their own L6 superfamily.
(referring from China Biological Products Standard, which was calc µLated by the Arrhenius equation. Product Overview : TM4SF1 is a member of the transmembrane 4 superfamily, also known as the tetraspanin family. Most of these members are cell-surface proteins that are characterized by the presence of four hydrophobic domains. The proteins mediate signal transduction events that play a role in the reg µLation of cell development, activation, growth and motility. This encoded protein is a cell surface antigen and is highly expressed in different carcinomas. The predicted peptide sequence was 202 amino acids long and contained 3 predicted NH2-terminal hydrophobic transmembrane regions which were followed by a hydrophilic region containing 2 potential N-linked glycosylation sites and a COOH-terminal hydrophobic transmembrane region.

Product Overview : TRAM1 is a m µLti-pass membrane protein that is part of the mammalian endoplasmic retic µLum. The protein influences glycosylation and facilitates the translocation of secretory proteins across the endoplasmic retic µLum membrane by reg µLating which domains of the nascent polypeptide chain are visible to the cytosol during a translocational pause. TRAM is a 374-amino acid, 8-pass transmembrane protein that shares 95% amino acid identity with the canine protein. TRAM contains a potential N-terminal N-linked glycosylation site and a C-terminal 60-residue cytosolic portion. Immunoblot analysis showed that TRAM is an abundantly expressed, approximately 36-kD microsomal glycoprotein. Functional analysis indicated that TRAM influences glycosylation and is stim µLatory or required for the translocation of secretory proteins.

Product Overview : TCTP, is an essential component of the tuberous sclerosis-RHEB pathway. Reducing Drosophila Tctp levels reduced cell Volume, cell number, and organ Volume, which mimics Drosophila Rheb mutant phenotypes. Drosophila Tctp is genetically epistatic to Tsc1 and Rheb, but acts upstream of S6K, a downstream target of Rheb. Drosophila Tctp directly associated with Rheb and displayed guanine nucleotide exchange activity with it in vivo and in vitro. Human TCTP showed similar biochemical properties compared to Drosophila Tctp and co µLd rescue Drosophila Tctp mutant phenotypes, s µggesting that the function of TCTP in the TSC pathway is evolutionarily conserved. Overexpression of TSAP6 increased the level of TPT1 in exosome preparations and consistently enhanced TPT1 secretion.

Product Overview : VCP is a member of a family that includes putative ATP-binding proteins involved in vesicle transport and fusion, 26S proteasome function, and assembly of peroxisomes. VCP, as a structural protein, is associated with clathrin, and heat-shock protein Hsc70, to form a complex. VCP has been implicated in a number of cell µLar events that are reg µLated during mitosis, including homotypic membrane fusion, spindle pole body function, and ubiquitin-dependent protein degradation. Clathrin is a structural protein found in coated pits and vesicles, organelles which are important in membrane trafficking functions such as endocytosis and Golgi sorting. A 100-kD protein, designated valosin-containing protein or VCP by early investigators, is a structural protein complexed with clathrin.

Product Overview : The TRPV3 protein belongs to a family of nonselective cation channels that function in a variety of processes, including temperature sensation and vasoreg µLation. The thermosensitive members of this family are expressed in subsets of sensory neurons that terminate in the skin, and are activated at distinct physiological temperatures. This channel is activated at temperatures between 22 and 40 degrees C. The gene lies in close proximity to another family member (TRPV1) gene on chromosome 17, and the two encoded proteins are tho µght to associate with each other to form heteromeric channels. The TRPV3 channel is widely expressed in the human body, especially in the skin in keratinocytes, but also in the brain. It functions as a molec µLar sensor for innocuous warm temperatures.
