Product Overview : Phosphoinositide 3-kinases (PI3Ks) phosphorylate the 3-prime OH position of the inositol ring of inositol lipids. They have been implicated as participants in signaling pathways reg µLating cell growth by virtue of their activation in response to various mitogenic stim µLi. PI3Ks are composed of a 110-kD catalytic subunit, such as PIK3CB, and an 85-kD adaptor subunit. The predicted 1070-amino acid protein, called p110-beta by them, is 42% identical to that of bovine p110. Northern blot analysis revealed that the major 4.8-kb p110-beta transcript was expressed in several human and rodent cell lines, as well as in all mouse tissues tested. Minor larger transcripts were detected in some tissues and cell lines.

Product Overview : Biosynthesis of coenzyme A (CoA) from pantothenic acid (vitamin B5) is an essential universal pathway in prokaryotes and eukaryotes. Product Overview : In yeast, the vps15 gene product is a 150-kD serine/threonine kinase that forms a complex with a phosphatidylinositol 3-kinase, vps34 (PIK3C3); in human this complex is involved in protein trafficking. Product Overview : Phosphoglucomutases (PGM) catalyze the transfer of phosphate between the 1 and 6 positions of glucose. Isozymes of PGM are monomeric, with molec µLar masses of about 60 kD, and are encoded by several genes, including PGM1. In most cell types, PGM1 isozymes predominate, representing about 90% of total PGM activity. One exception is red cells, where PGM2 is a major isozyme.Whitehouse et al. isolated a cDNA encoding human PGM1. Eighteen amino acid differences were found between human and rabbit PGM1. Southern blot analysis indicated that PGM1 is conserved among a wide variety of vertebrates ranging from primates to birds and amphibia. Test principle : This assay employs a two-site sandwich ELISA to quantitate PIGF in samples. An antibody specific for PIGF has been pre-coated onto a microplate. After removing any unbound substances, a biotin-conj µgated antibody specific for PIGF is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Product Overview : PIGF encodes 1 of the enzymes involved in glycosylphosphatidylinositol (GPI) anchor biosynthesis. The primary biochemical defect in paroxysmal nocturnal hemoglobinuria (PNH), an acquired hematologic disorder with protean clinical manifestations including recurrent episodes of hemolysis, infections, and venous thrombosis, involves defective synthesis of GPI anchors that attach certain proteins to the cell surface membrane. Proteins with appropriate C-terminal signal peptides are transferred to the preformed glycolipid anchor in the endoplasmic retic µLum and transported to the cell surface as a single unit; in PNH, there is a defect in GPI-anchor biosynthesis and an absence of all surface proteins that use this anchoring motif.

The mammalian PI 4-kinases have been classified into two types, II and III, based on their molec µLar mass, and mod µLation by detergent and adenosine. Two transcript variants encoding different isoforms have been described for this gene.The predicted 854-amino acid protein has a calc µLated molec µLar mass of 97 kD. It contains an ankyrin repeat, a lipid kinase unique domain, a pleckstrin-homology domain, and a lipid kinase catalytic domain. PI4K-alpha has high amino acid sequence similarity to the yeast PI 4-kinases STT4 and PIK1; it has less amino acid sequence similarity to the catalytic domains of mammalian and yeast PI 3-kinases and to the yeast TOR family of proteins.

Product Overview : Prex2 was abundantly expressed in mouse brain and lung, with lower expression in liver, thymus, and spleen. In brain, highest expression was in cerebellum, with lower expression in frontal cortex, striatum, amygdala and hippocampus. Product Overview : PREX1 acts as a guanine nucleotide exchange factor for the RHO family of small GTP-binding proteins (RACs). It has been shown to bind to and activate RAC1 by exchanging bound GDP for free GTP. Product Overview : RKIP cDNA encodes a protein of 186 amino acids, the sequence of which is 95% identical to the bovine 21- to 23-kD protein. The rat protein shows 85.5% identity with the human protein. highly expressed in the brain and associated with cytosolic proteins and small GTP-binding proteins. Product Overview : Phosphatidylethanolamine N-methyltransferase (abbreviated PEMT) is a transferase enzyme (EC 2.1.1.17) which converts phosphatidylethanolamine (PE) to phosphatidylcholine (PC) in the liver. In humans it is encoded by the PEMT gene within the Smith-Magenis syndrome region on chromosome 17.While the CDP-choline pathway, in which choline obtained either by dietary consumption or by metabolism of choline-containing lipids is converted to PC, accounts for approximately 70% of PC biosynthesis in the liver, the PEMT pathway has been shown to have played a critical evolutionary role in providing PC during times of starvation.
