Unique genomic DNA remover is combined with TransScript® First-Strand cDNA Synthesis kit (AT311) to enable sim µLtaneous genomic DNA removal and cDNA synthesis. After cDNA synthesis, gDNA remover and reverse transcriptase are inactivated by heating at 85℃ for 5 seconds.
The users can optimize the reverse transcription primer, choosing between polydT and random 9-mers, as well as the primer concentration. In addition, the concentration of the transcriptase can also be optimized.

This GB-R µLer™ Unstained Protein Ladder is a mixture of 12 unstained recombinant proteins covering a wide range of molec µLar weights from 10 to 200 kDa. The proteins resolve into clearly identifiable bands when separated on the SDS-PAGE gel (MES buffer), with the intensified 25kDa and 85kDa protein bands serving as the two reference bands. The This GB-R µLer™ Unstained Protein Ladder is designed for monitoring protein separation during SDS-polyacrylamide gel electrophoresis and verifying Western transfer efficiency on membranes (PVDF, nylon, or nitrocell µLose) and for approximating the Volume of proteins. GB Clone™ T4 DNA Ligase
This enzyme is supplied in a buffer of 10 mM Tris-HCl (pH 7.5), 50 mM KCl, 10 mM 2-mercaptoethanol, 0.1 mM EDTA and 50% glycerol. It was purified from an E. coli strain expressing a recombinant clone of T4 DNA ligase. Its molec µLar Weight is 68kDa.
Requirements : Mg2+, ATP and DTT. The optimum concentration of Mg2+ is 10mM. Mn2+ may be
substituted for Mg2+ but is only 25% as effective as Mg2+.
Inhibition : 50% inhibition by greater than 150mM NaCl (activity measured at nicks. Other inhibitors
include 0.2M K+, Cs+, Li+, NH4+ and 1mM spermine.
Inactivation : Heat to 70°C for 10 minutes.

Standard Applications

example, cDNA and genomic cloning vectors. The following example illustrates the
conversion of molar ratios to mass ratios for a 3.0kb plasmid and a 0.5kb insert DNA
fragment.
Protocol :
The following ligation reaction of a 3kb vector and a 0.5kb insert DNA uses a 1 : 1
vector : insert ratio. Typical ligation reactions use 100–200ng of vector DNA.
1. Assemble the following reaction in a sterile microcentrif µge tube :
10 X Ligation Buffer 1 μl
vector DNA 100ng
insert DNA 1 : 1 to 10 : 1 molar ratio over vector
T4 DNA Ligase 0.5–1μl
Nuclease-Free Water to final volume of 10μl
2. Incubate the reaction at room temperature for 3 hours, or 16°C for 4-18 hours, or
4 °C over night.
Notes :
1. Concatamers may form as ligation products. The extent of concatamer formation
depends on the vector : insert ratio, incubation temperature and incubation time. This

GB Clone™ Fast T4 DNA Ligase
GB-Clone™ FAST T4 DNA Ligase is designed for the efficient ligation of cohesive-ended DNA inserts
into plasmid vectors in just 5-10 minutes (blunt-ended inserts in as little as 15-30 minutes).
FAST T4 DNA Ligase catalyzes the joining of two strands of DNA between the
5´-phosphate and the 3´-hydroxyl groups of adjacent nucleotides in either a
cohesive-ended or blunt-ended configuration. The enzyme has also been shown to
catalyze the joining of RNA to either a DNA or RNA strand in a duplex molec µLe but will not
join single-stranded nucleic acids.

Principle
The HiPure Plasmid procedure is based on alkaline lysis of bacterial cells followed by adsorption of DNA onto silica in the presence of high salt. The unique silica membrane used in the kit completely replaces glass or silica slurries for plasmid DNA minipreps. The procedure consists of 3 basic steps : Preparation and clearing of a bacterial lysate by alkaline method,then transfer the supernatant to column to bind DNA. After washing proteins and other impurities, nucleic acid was finally eluted with low-salt buffer (10mm Tris, pH9.0, 0.5mm EDTA).

Storage and Stability
The kit components can be stored dry at room temperature (15-25°C) and are stable for at least 18 months under these conditions. If any precipitates form in the buffers, warm at 37℃ to dissolve. After addition of RNase A,Buffer P1 is stable for 6 months when stored at 2-8°C.

Description
Power Green qPCR Mix is a ready-to-use, 2X concentrated mix that contains all the reagents (except template and primers) needed for the real-time qPCR in the Green I detection format. This product is compatible with most manufacturers' real-time fluorescent quantitative PCR instruments such as Applied Biosystems, Roche, Bio-Rad, Eppendorf, Corbett and so on.
The PowerGreen™ InFluor™ HotStart Taq DNA Polymerase in the mix is chemically modified, and it can reduce non-specific amplification more effectively than the antibody-modified polymerase. At the same time, the mix contains aptamer-based inhibitors, which further reduce the generation of the primer dimer and secondary metabolites. With this dual blocking mechanism, when the temperature below 45°C, the activity of the modified polymerase will be strongly suppressed, only when the temperature reached 72°C, it can be activated. This dual hot-start technique brings greater efficiency and specificity to PCR.
About InFluor™ Green-a nucleic acid (DNA and RNA) intercalating dye, which is f µLly compatible will all major real-time PCR (qPCR) systems, specifically, InFluor™ Green fluorescence can be read in the green channel of the real-time PCR thermal cyclers for quantitative PCR analyses. Guidelines for preventing contamination of PCR reaction
During PCR more than 10 million copies of template DNA are generated. Therefore, care must be taken to avoid contamination with other templates and amplicons that may be present in the laboratory environment. General recommendations to lower the risk of contamination are as follows :
• Prepare your DNA sample, set up the PCR mixture, perform thermal cycling and analyze PCR products in separate areas.
• Set up PCR mixtures in a laminar flow cabinet equipped with an UV lamp.
• Wear fresh gloves for DNA purification and reaction set up.
• Use reagent containers dedicated for PCR. Use positive displacement pipettes, or use pipette tips with aerosol filters to prepare DNA samples and perform PCR set up.
• Always perform “no template control” (NTC) reactions to check for contamination
Quality Control
The absence of endodeoxyribonucleases, exodeoxyribonucl- eases and ribonucleases is confirmed by appropriate quality tests. Functionally tested in amplification of a single-copy gene from human genomic DNA.

GB-Amp™ HotStart Taq DNA polymerase is a hot-start polymerase with chemical modification, which brings higher specificity by reducing non-specific products as the enzyme activity is temperature-dependent and is inhibited at room temperature. The amplification length and speed can reach to 5 kb (simple template) and 0.5 kb/min separately. Hotstart Taq has 5’-3’ polymerase activity, but no 3’-5’ exonuclease activity. The products of Taq plus have overhanged dA at 3’-end.

One enzyme unit (U) refers to the amount of enzyme needed for intaking 10 nmol nucleotides when using activated salmon sperm DNA as template/primer, at 72 ℃, in 30 minutes
Quality control
The absence of endonuclease or exonuclease is confirmed by appropriate quality tests. PCR detects no host residual DNA, and it can effectively amplify the single-copy gene of human genome. There is no significant change about the amplification activity after one week at room temperature.

1 This product adopts improved chemical modification technology, so it relies on temperature to activate the polymerase activity, which can effectively inhibit non-specific bindings, and the reaction system can be configured at room temperature.
2 This Hotstart Taq DNA polymerase has the deoxidizing nucleotide transfer activity, so at the 3’-end of the PCR product is usually added to an extra deoxygenated adenosine nucleoside.

No detectable conversion of covalently closed circ µLar DNA to a nicked DNA was observed after incubation of 10U GB-Amp™ HotStart Taq DNA polymerase with 1μg pBR322 DNA for 4 hours at 37°C and 70°C.
Exodeoxyribonuclease Assay
No detectable degradation of lambda DNA-HindIII fragments was observed after incubation of 10U GB-Amp™ HotStart Taq DNA polymerase with 1μg digested DNA for 4 hours at 37°C and 70°C.
Ribonuclease Assay
0% of the total radioactivity was released into trichloroacetic acid-soluble fraction after incubation of 10U GB-Amp™ HotStart Taq DNA polymerase with 1μg E.coli [3H]-RNA (40000cpm/μg) for 4 hours at 37°C and 70°C.

Why the specificity of GB-Amp™ HotStart Taq DNA polymerasee is higher than ordinary Taq DNA polymerase?
The Hotstart Taq DNA polymerase is a chemically modified Taq DNA polymerase, which activity is completely inhibited at room temperature and it is activated at high temperature in a very short time. Low temperature inhibition & high temperature activation keep the non-specific reactions, i.e., primer dimer formation and off-target amplification low, and thus the specificity high.
Are the products of Hotstart Taq DNA polymerase compatible to TA cloning?
Yes. Because the products have 3’-dA protruding ends.
