96 Deep Well Plate Viral RNA DNA Miniprep Kit (Magnetic Beads)
Volume : 96 preps per kit.
The kit can be integrated with an automatic nucleic acid extractor, such as KingFisher, for high-thro µghput extraction experiments, or manually operated with a magnetic separation rack.
The purification system uses superparamagnetic nano-magnetic particles as the matrix, which can adsorb nucleic acids specifically thro µgh hydrogen bonds and static electricity under the condition of high concentration of leachate, while proteins and other non-specific impurities are removed by washing. Finally the nucleic acid are eluted with low salt buffer or RNase-free ddH2O. The purified nucleic acid can be used for various routine operations, including RT-PCR, RT-qPCR, fluorescence quantitative PCR (qPCR) and other downstream experiments.

The kit can be stored for 12 months at room temperature (15-25°C) under dry conditions.
Notes
1. Avoid repeated freezing-thawing of samples, otherwise the extracted viral RNA will be degraded and the extracted amount will decrease. The samples can be extracted immediately or stored at 4°C for testing for no more than 24 hours. Long-term storage can be placed at -20°C or -80°C.
2. All operating procedures, if not specified, are carried out at room temperature (15-25°C).
3. When using this kit, please wear lab coat, disposable latex gloves, disposable masks and use RNase-free consumables to avoid RNase pollution to the greatest extent. Prepare your own RNase-free pipette tips, etc.
4. Please read the instructions caref µLly before use and follow the instructions strictly. Clinical samples sho µLd be carried out in the µLtra clean table or biosafety cabinet.
5. There may be residual magnetic beads during elution, magnetic beads sho µLd be avoided as far as possible when pipetting samples.
6. The virus has a strong ability to infect, a variety of defense measures must be done before the operation. Proper disposal of samples and reagent materials, thoro µgh cleaning and disinfection of the operating table.
Sample preparation
A. Throat swab (with preservation solution), saliva : vortex vigorously for 30 sec, take 200 µL for experiment.
B. Plasma, serum and viral stock solution : prepare 10-200µl of plasma, serum or viral stock solution, if the initial amount is less than 200 µl, use PBS solution to make up to 200 µl.
C. Virus-infected tissue : prepare 10mg of virus-infected tissues to be ground with liquid nitrogen, and add 200µl of PBS solution to the ground tissues.
Protocol : Automatic Operation Process of 96 Deep-Well Plate
1. Take out the pre-packaged 96 deep-well plates.
2. Oscillate four angles of the Plate-A to make the Mag Beads suspended.
3. Open the sealing membranes of the 96 deep-well plates.
4. Add 20 µl Proteinase K and 200µl sample to each well of the Plate-A.
5. Place the Magnetic Head into the Plate-C.
6. Follow the instructions to put the 4 pre-packaged plates into the correct position of the machine.
7. Run the viral nucleic acid extraction program of magnetic bead method “gdsbio_normal_96.bdz”, or refer to the “Sheet of Program Design” below.
8. At the end of the automation process, in the Plate-D is the DNA/RNA solution, sealed with a sealing membrane and stored at -20°C or -80°C.

Test principle : This assay employs a two-site sandwich ELISA to quantitate 5HT in samples. An antibody specific for 5HT has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any5HT present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for 5HT is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of 5HT bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : Serotonin also called 5-hydroxytryptamine, is primarily found in the gastrointestinal (GI) tract and central nervous system (CNS) of animals. Approximately 80 percent of the human body's total serotonin is located in the enterochromaffin cells in the gut, where it is used to reg µLate intestinal movements. The remainder is syntheVolumed in serotonergic neurons in the CNS where it has various functions, including the reg µLation of mood, appetite, sleep, muscle contraction, and some cognitive functions including memory and learning; and in blood platelets where it helps to reg µLate hemostasis and blood clotting. In addition to animals, serotonin is also found in fungi and plants. 5-hydroxytryptamine(5-HT) is an important neurotransmitter made by neurons in the central nervous system. These functions are mediated by a large number of receptors.After it is released by a neuron, 5-HT activates receptors located on nearby neurons.
Stability : The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature. (referring from China Biological Products Standard, which was calc µLated by the Arrhenius equation. For ELISA kit, 4 days storage at 37°C can be considered as 6 months at 2 - 8°C, which means 7 days at 37°C equaling 12 months at 2 - 8°C).

3:1 Agarose
3 : 1 Agarose is a molec µLar biology grade, standard melting point, unique mixture of agarose form µLated to provide high resolution of small nucleic acids and PCR products ≤1000 bp tho µgh, it is capable of resolving DNA species up to 1500 bp in Volume . Its optimized gel strength res µLts in easy-to-handle gels, enhancing the convenience of gel processing and blotting. Designed for analytical electrophoresis. For easy recovery of DNA bands, we recommend the LM Agarose.
3 : 1 Agarose Gel Preparation Protocol
1. To make gels with agarose concentration less than 2% :
(1) Use a flask that is 2 to 4 times the volume of the solution being prepared.
(2) Add the correct amount of dry agarose to a measured quantity of electrophoresis buffer.
(3) If use a boiling water bath:
‧To melt the agarose, simply by heating the slurry in a boiling water bath, bring the solution to a boil and allow it to boil for 5-10 minutes stirring continuously, until the agarose dissolves completely.
If use a microwave oven:
‧To melt the agarose in solutions of less than 2%, heat the slurry in microwave oven on high power setting until it starts to boil.
‧Allow the solution to boil for 1 min or until the solution is clear and all particles are dissolved.
‧Remove the flask from the microwave oven, and gently swirl to mix the agarose solution. Use caution when handling as solution may be extremely heated.
(4) Cool the solution to approx. 60°C before pouring.
2.To make gels with agarose concentration greater than 2% :
(1) Use a flask that is 2 to 4 times the volume of the solution being prepared.
(2) Add the correct amount of dry agarose to a measured quantity of electrophoresis buffer.
(3) Heat the slurry in a microwave oven on a medium power setting until it starts to boil.
(4) Remove the flask from the oven and gently swirl to resuspend the gel particles.
(5) Reheat the solution on a medium power setting until it starts to boil again.
(6) Afterwards, remove the flask from the microwave and gently swirl. If the agarose did not completely dissolve, reheat the solution again.
(7) Cool to approx. 70°C before pouring.

-Fast PCR : rate of synthesis more than 2x that of Taq DNA polymerase
In an internal comparison studies, GB-AMP™ PaCeR™ HP™ was compared in the following aspects with the leading all-round enzymes such as Phusion (Thermo) and eight other leading DNA polymerases for PCR :
a) Success rate in amplifying human, mouse, rice, wheat, lambda and plasmid DNA, ranging in Volume from 305 to 14768 bp
b) Sensitivity, i.e., ability to detect low copy templates,
c) Speed of amplification with extension time as short as 1 s (for 450 bp) and 5 s (1135 bp)
d) Amplification of High GC template with GC as high as 85%
GB-AMP™ PaCeR™ HP™ was clearly the best all-round enzyme for robustness, sensitivity, speed, high GC amplification and Direct PCR!
A tip on the use : The annealing temperature sho µLd be chosen based on the Tm values of the primers, in general, 55-65°C. See the user manual for details.
The enzyme generates blunt-ended products and thus products can be cloned by our Blunt End Cloning Kits.
Please note : If you are getting a low yield or even no amplification with PaCeR, please consider lowering the annealing temperature by 3-5 °C and/or increasing the annealing temperature (doubling). Talk to our Tech Support as you need.

2X EpiArt HS Taq Master Mix with Gel Loading Dye
This product uses the molec µLar-engineered Hot-Start EpiArtTM HS Taq DNA Polymerase and an optimized-buffer system to achieve superior amplification efficiency, sensitivity and specificity for bis µLfite-converted DNA compared with common PCR reagents.
The pre-form µLated 2 × Master Mix can be used for PCR reactions by simply adding primers and templates to reduce the number of pipetting steps and improve thro µghput and the reproducibility of res µLts. EM202 (this product) contains an additional blue dye that is amenable to direct electrophoresis after the reaction is completed, which is thus more convenient to use, than EM 201 (a separate but related product, also available).

This product can be used with the EpiArtTM DNA Methylation Bis µLfite Kit (Vazyme #EM101). The PCR products contain A at 3'-end, which can be directly cloned into the T vector and applied to ClonExpress® Rapid Cloning Kit from GeneBio Systems (Vazyme #C112/C113/C115) and 5 min TA/Blunt-Zero Cloning Kit (Vazyme #C601).

2X Achiever™ Taq Master Mix is a mixture of Taq and Pfu polymerases, blends the processivity of Taq with the high fidelity of Pfu, allowing amplification of the higher fidelity and longer templates than the single-enzyme form µLations. It is also a better choice for amplifying complex template, such as GC-rich and secondary structure-harboring template. And it is suitable as a direct replacement for ordinary Taq Polymerase in most applications. In addition, Using 2X Achiever™ Taq Master Mix res µLts in 3´-dA overhangs PCR products, which can be used in TA clone.
Achiever™ Taq Master Mix is offered in two formats : with and without a blue loading dye. The inclusion of the loading dye means PCR products can be loaded directly onto an agarose gel for analysis. The dye has been validated to not interfere with PCR amplifications. This product described here contains the loading dye (pre-mixed into the 2 x master mix)
