Test principle : This assay employs a two-site sandwich ELISA to quantitate TMEM173 in samples. An antibody specific for TMEM173 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTMEM173 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for TMEM173 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TMEM173 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : The deduced 379-amino acid protein has a calc µLated molec µLar mass of 42.2 kD. It has 5 putative N-terminal transmembrane domains, a signal cleavage site in the first transmembrane domain, and a leucine-rich region that overlaps the first 4 transmembrane domains. Northern blot analysis detected STING expression in all tissues examined. Confocal microscopy and fractionation analysis of human embryonic kidney 293 cells revealed that STING predominantly associated with the endoplasmic retic µLum (ER). Western blot analysis of 293 cells detected endogenous STING at an apparent molec µLar mass of 42 kD.STING activated both the NF-kappa-B and IRF3 transcription pathways to induce expression of IFN-alpha (IFNA1) and IFN-beta and exert a potent antiviral effect.
Stability : The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature. (referring from China Biological Products Standard, which was calc µLated by the Arrhenius equation. For ELISA kit, 4 days storage at 37°C can be considered as 6 months at 2 - 8°C, which means 7 days at 37°C equaling 12 months at 2 - 8°C).

Product Overview : TMEM171 (transmembrane protein 171) is a 324 amino acid protein encoded by a gene mapping to human chromosome 5. With 181 million base pairs encoding around 1000 genes, chromosome 5 is about 6% of human genomic DNA. It is associated with Cockayne syndrome thro µgh the ERCC8 gene and familial adenomatous polyposis thro µgh the adenomatous polyposis coli (APC) tumor suppressor gene. Treacher Collins syndrome is also chromosome 5 associated and is caused by insertions or deletions within the TCOF1 gene. Deletion of the p arm of chromosome 5 leads to Cri du chat syndrome. Deletion of 5q or chromosome 5 altogether is common in therapy-related acute myelogenous leukemias and myelodysplastic syndrome.

Product Overview : TMEM169 Evidence at transcript level. In plain language, a transmembrane protein (TP) is a protein that goes from one side of a membrane thro µgh to the other side of the membrane. Many TPs function as gateways or "loading docks" to deny or permit the transport of specific substances across the biological membrane, to get into the cell, or out of the cell as in the case of waste byproducts. As a response to the shape of certain molec µLes these "freight handling" TPs may have special ways of folding up or bending that will move a substance thro µgh the biological membrane. A transmembrane protein is a polytopic protein that spans an entire biological membrane. Transmembrane proteins aggregate and precipitate in water. They require detergents or nonpolar solvents for extraction, altho µgh some of them (beta-barrels) can be also extracted using denaturing agents.

Product Overview : Constitutive activation of the Ras pathway triggers an irreversible proliferation arrest reminiscent of replicative senescence. Transcription of this gene is upreg µLated in response to activation of the Ras pathway, but not under other conditions that induce senescence. RIS1 is similar to a rat cell surface receptor proposed to function in a neuronal survival pathway. RIS1 is not upreg µLated in association to any of the above-mentioned processes, but exclusively during Ras-senescence. Furthermore, RIS1 is also upreg µLated by the transcriptional factor Ets2, which is a known mediator of Ras-induced senescence. Interestingly, RIS1 is located at chromosomal position 3p21.3 and, more specifically, it is included in a short segment of just 1 Mb previously defined by other investigators for its tumor-suppressor activity.

Product Overview : TMED9 Belongs to the EMP24/GP25L family.Contains 1 GOLD domain. Members of the p24 (p24/gp25L/emp24/Erp) family of proteins have been shown to be critical components of the coated vesicles that are involved in the transportation of cargo molec µLes from the endoplasmic retic µLum to the Golgi complex. The p24 proteins form hetero-oligomeric complexes and are believed to function as receptors for specific secretory cargo. The identification of the GOLD domain co µLd aid in directed investigation of the role of the p24 proteins in the secretion process. The newly detected group of GOLD-domain proteins, which might sim µLtaneously bind membranes and other proteins, point to the existence of a novel class of adaptors that co µLd have a role in the assembly of membrane-associated complexes or in reg µLating assembly of cargo into membranous vesicles.

Standards and samples are pipetted into the wells and anyTM6SF1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for TM6SF1 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TM6SF1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : TM9SF1 shows a large N-terminal hydrophilic part and a C-terminal part with nine putative hydrophobic regions characteristic of integral transmembrane domains. Computer searches with sequence databases revealed homologies with three complete yeast proteins and with at least 19 human, 10 plant and one nematode short unidentified protein sequences translated from Expressed Sequence Tags (ESTs). Remarkably, this hMP70 protein retains between 27 and 31% overall sequence identity with the yeast proteins.Gene expression of hMP70 appears to be ubiquitous, as the mRNA is detectable in all human tissues analysed so far, as shown by Northern blot analysis. Furthermore, a protein of about 70 kDa is detectable in different mammalian cell lines, as shown by immunoblot analysis.

Product Overview : Like other TM4SF proteins, TM4SF5 is cysteine-rich. TM4SF5 shares 50% amino acid identity with the tumor-associated antigen L6 (TM4SF1). Northern blot analysis of various normal human tissues, namely intestine, pancreas, stomach, colon, esophagus, bladder, uterus, placenta, and adipose tissue, detected TM4SF5 expression only in intestine and 1 of 14 pancreas samples. TM4SF5 was overexpressed in 13 of 14 pancreatic adenocarcinomas, as compared with both normal pancreatic and chronic pancreatitis tissues. TM4SF5 expression was also found in 2 of 2 gastric cancers, 1 of 2 colon carcinomas, 1 of 1 carcinoma of the papilla vateri, and 1 of 1 soft-tissue sarcoma. TM4SF5 was expressed as an approximately 1-kb transcript in the pancreatic tumors.

Product Overview : TGF-? is capable of producing a variety of effects and virtually all cell types respond to this factor in some way. The inappropriate presence of active TGF-?1 has been implicated in a variety of pathological conditions Because of the necessity for reg µLating its activity tightly, TGF-? is secreted by cells in the form of an inactive complex. This complex consists of TGF-?1 associated non-covalently with a protein designated the latency associated peptide (LAP). TGF-?1 and LAP represent components of a pro-peptide that is cleaved in a post-golgi compartment prior to secretion. LAP and TGF-?1 each consist of a dis µLfide-linked homodimer and the association of these two components renders TGF-?1 inactive and inaccessible to anti-TGF-? antibodies.

Product Overview : The transcriptional coactivator CBP/p300 associates with thro µgh 2 sites, an N-terminal domain that interacts with the C-terminal region of unphosphorylated p65, and a second domain that only interacts with p65 phosphorylated on serine-276. Phosphorylation by PKA both weakens the interaction between the N- and C-terminal regions of p65 and creates an additional site for interaction with CBP/p300. Therefore, PKA reg µLates the transcriptional activity of NF-kappa-B by mod µLating its interaction with CBP/p300.The p50 (NFKB1)/p65 (RELA) heterodimer is the most abundant form of NFKB. The NFKB complex is inhibited by I-kappa-B proteins (NFKBIA or NFKBIB ), which inactivate NFKB by trapping it in the cytoplasm.

Product Overview : The deduced 368-amino acid TRa protein contains 2 follistatin-like domains and a conserved EGF-like domain. It also has a predicted N-terminal signal peptide, a transmembrane domain, a cytosolic tail G protein-activating motif, and potential sites for N-linked glycosylation and glycosaminoglycan attachment. Unlike other EGF/NRG family members, TRa has a histidine replacing a conserved arginine essential for EGF receptor recognition. The TRb and TRc proteins contain 418 and 379 amino acids, respectively, and differ from one another and TRa in the cytoplasmic domain. Northern blot analysis detected expression of approximately 2.2- and 3.1-kb TR transcripts predominantly in brain. The 374-amino acid TMEFF2 protein shares approximately 36% and 99% sequence identity with TMEFF1 and mouse Tmeff2, respectively.

Product Overview : TLR 4 is a member of the Toll-like receptor (TLR) family which plays a fundamental role in pathogen recognition and activation of innate immunity. TLRs are highly conserved from Drosophila to humans and share structural and functional similarities. They recognize pathogen-associated molec µLar patterns (PAMPs) that are expressed on infectious agents, and mediate the production of cytokines necessary for the development of effective immunity. The various TLRs exhibit different patterns of expression. This receptor is most abundantly expressed in placenta, and in myelomonocytic subpop µLation of the leukocytes. It has been implicated in signal transduction events induced by lipopolysaccharide (LPS) found in most gram-negative bacteria. Mutations in this gene have been associated with differences in LPS responsiveness.
Product Overview : TLR 3 is a member of the Toll-like receptor family of pattern recognition receptors of the innate immune system. Discovered in 2001TLR3 recognizes double-stranded RNA, a form of genetic information carried by some viruses such as reoviruses. Upon recognition, TLR 3 induces the activation of NF-kB to increase production of type I interferons which signal other cells to increase their antiviral defenses. Double-stranded RNA is also recognised by the cytoplasmic receptors RIG-I and MDA-5.The structure of TLR3 was reported in June 2005 by researchers at The Scripps Research Institute. TLR3 forms a large horseshoe shape that contacts with a neighboring horseshoe, forming a "dimer" of two horseshoes.

Recovery : 1.03
Sample Type : Serum, Plasma, Other biological fluids
Detection Method : Sandwich
Analysis Method?? : Quantitive
Test principle : This assay employs a two-site sandwich ELISA to quantitate TLR10 in samples. An antibody specific for TLR10 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTLR10 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for TLR10 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TLR10 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : Toll-like receptor 10 is a member of the Toll-like receptor (TLR) family which plays a fundamental role in pathogen recognition and activation of innate immunity. This gene is most highly expressed in lymphoid tissues such as spleen, lymph node, thymus, and tonsil. Its exact function is not known. M µLtiple alternatively spliced transcript variants encoding the same protein have been found for this gene.
Test principle : This assay employs a two-site sandwich ELISA to quantitate TOLLIP in samples. An antibody specific for TOLLIP has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTOLLIP present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for TOLLIP is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TOLLIP bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : Toll interacting protein, also known as TOLLIP, is an inhibitory adaptor protein .It is an inhibitory adaptor protein within Toll-like receptors (TLR). The TLR pathway is a part of the innate immune system that recognizes structurally conserved molec µLar patterns of microbial pathogens, leading to an inflammatory immune response. Compared with the f µLl-length isoform, 2 of the human proteins lack the N-terminal TBD, and 1 lacks the C2 domain. A mouse-specific isoform lacks the C-terminal CUE domain. EST database analysis s µggested that f µLl-length human TOLLIP is ubiquitously expressed. Expression of the other human variants appeared to be more restricted, with evidence for enrichment in brain.
Product Overview : TM2D2 contains a structural mod µLe related to that of the seven transmembrane domain G protein-coupled receptor superfamily. This protein has sequence and structural similarities to the beta-amyloid binding protein (BBP), but, unlike BBP, it does not reg µLate a response to beta-amyloid peptide. This protein may have reg µLatory roles in cell death or proliferation signal cascades. This gene has m µLtiple alternatively spliced transcript variants which encode two different isoforms. The deduced 214-amino acid protein contains several features of a G protein-coupled receptor, including 2 putative transmembrane domains in its C-terminal half, a DRY motif, and conserved cysteines and lysines. Northern blot analysis detected variable expression of a 1.35-kb BLP1 transcript in all human tissues examined.

The color development is stopped and the intensity of the color is measured.
Product Overview : Leco et al. (1997) cloned cDNAs encoding mouse Timp4. The predicted Timp4 protein contains the hallmarks of TIMP proteins, including 12 conserved cysteine residues and 2 short conserved sequence motifs. Northern blot analysis of ad µLt mouse tissues detected Timp4 expression in brain, heart, ovary, and skeletal muscle. Olson et al., 1998 determined that the TIMP4 gene contains 5 exons that span 6 kb of genomic DNA. They demonstrated a high degree of conservation of gene structure in the TIMP family. By FISH, Olson et al., 1998 mapped the TIMP4 gene to chromosome 3p25. By interspecific backcross analysis, they mapped the mouse Timp4 gene to chromosome 6 in a region of syntenic homology with human chromosome 3.
