Test principle : This assay employs a two-site sandwich ELISA to quantitate DPAGT1 in samples. An antibody specific for DPAGT1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyDPAGT1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for DPAGT1 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of DPAGT1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : DPAGT1 is an enzyme that catalyzes the first step in the dolichol-linked oligosaccharide pathway for glycoprotein biosynthesis. This enzyme belongs to the glycosyltransferase family 4. This protein is an integral membrane protein of the endoplasmic retic µLum. The congenital disorder of glycosylation type Ij is caused by mutation in the gene encoding this enzyme. The cDNA encodes a deduced 400-amino acid protein with a calc µLated molec µLar mass of 44.7 kD. DPAGT1 contains an N-terminal signal peptide, 2 potential dolichol-binding sequences, and 4 sites for N-glycosylation. It shares 93% amino acid homology with hamster Dpagt, including 100% identity in the dolichol-binding region, and 42% homology with S. cerevisiae GlcNAc-1-P transferase.

Product Overview : UDP-glucuronate decarboxylase ( µgD; EC 4.1.1.35) catalyzes the formation of UDP-xylose from UDP-glucuronate. UDP-xylose is then used to initiate glycosaminoglycan biosynthesis on the core protein of proteoglycans. The topology of the deduced 420-amino acid protein was consistent with a type II transmembrane protein. Northern blot analysis of rat tissues detected a single µgD transcript with highest levels in heart, brain, and testes. Substantial levels were also detected in kidney, liver and lung, and lower levels in spleen and skeletal muscle. Western blot analysis demonstrated highest levels of the protein in kidney, liver, and brain, with negligible staining in heart, spleen, skeletal muscle, lung, and testes. Subcell µLar studies and histochemistry localized the protein to the perinuclear Golgi.

Product Overview : Glycosphingolipids (GSLs) are a group of membrane components that contain lipid and s µgar moieties. UDP-glucose ceramide glucosyltransferase ( µgCG) catalyzes the first glycosylation step in GSL biosynthesis. The product, glucosylceramide, is the core structure of more than 300 GSLs. Ichikawa et al. cloned a µgCG cDNA by expressing a human melanoma cell cDNA library in a µgCG-deficient cell line and screening for µgCG complementation. The predicted 394-amino acid µgCG protein has a potential signal-anchor sequence and a single transmembrane domain near the N terminus, and very hydrophobic regions close to the C terminus, which may interact with the membrane.

Product Overview : Ubiquitin-protein ligase E3A (UBE3A) is an enzyme that in humans is encoded by the UBE3A gene. This enzyme is involved in targeting proteins for degradation within cells. Protein degradation is a normal process that removes damaged or unnecessary proteins and helps maintain the normal functions of cells. Ubiquitin protein ligase 3A attaches a small protein called ubiquitin to proteins that sho µLd be degraded. Cell µLar structures called proteasomes recognize and digest proteins tagged with ubiquitin. Both copies of the UBE3A gene are active in most of the body's tissues. In the brain, however, only the copy inherited from a person's mother (the maternal copy) is normally active. The UBE3A gene is located on the long (q) arm of chromosome 15 between positions 11 and 13, from base pair 23,133,488 to base pair 23,235,220.

Product Overview : ATG7 was identified based on homology to Pichia pastoris GSA7 and Saccharomyces cerevisiae APG7. In the yeast, the protein appears to be required for fusion of peroxisomal and vac µLuolar membranes. The protein shows homology to the ATP-binding and catalytic sites of the E1 ubiquitin activating enzymes. The deduced 703-amino acid protein contains a central putative E1-like ATP-binding site (GxGxxG), conserved charged amino acids flanking the GxGxxG motif, and a putative E1 active site with a conserved catalytic cysteine. APG7L shares similarity with the E1 enzymes UBA2 and UBA3 (UBE1C), and it shares 38% identity with yeast Apg7. EST database analysis indicated that APG7L is expressed by many diverse tissues.

Product Overview : HIP2 belongs to the ubiquitin-conj µgating enzyme family. It binds selectively to a large region at the N terminus of huntingtin. Product Overview : Ubiquitin is a protein encoded by the UBB gene.one of the most conserved proteins known. Ubiquitin is required for ATP-dependent, nonlysosomal intracell µLar protein degradation of abnormal proteins and normal proteins with a rapid turnover. Ubiquitin is covalently bound to proteins to be degraded, and presumably labels these proteins for degradation. Ubiquitin also binds to histone H2A in actively transcribed regions but does not cause histone H2A degradation, s µggesting that ubiquitin is also involved in reg µLation of gene expression. This gene consists of three direct repeats of the ubiquitin coding sequence with no spacer sequence. Product Overview : Ubiquilin-2 is a ubiquitin-like protein (ubiquilin) that shares high degree of similarity with related products in yeast, rat and frog. Ubiquilins contain a N-terminal ubiquitin-like domain and a C-terminal ubiquitin-associated domain.
