Product Overview : SLC39A4 gene as a candidate intestinal zinc-specific transporter within the susceptibility locus for the zinc-deficiency type of acrodermatitis enteropathica (AEZ) on 8q24.3. The gene is expressed as an mRNA of 2.5 kb in only a few tissues, i.e., kidney, colon, duodenum and jejunum. The duodenum and jejunum are crucial sites of zinc absorption. These transcripts encode proteins of 647 and 622 amino acids sharing the same C-terminal 583 residues. Computational analysis predicted that the larger protein has a cytoplasmic membrane location and a secondary structure characteristic of a ZIP protein, consisting of 8 transmembrane domains organized in 2 blocks of 3 and 5, respectively. These 2 blocks are separated by a variable intracytoplasmic metal-binding site.

Product Overview : By screening a human temporal cortex cDNA library with a mouse zinc transporter-3 (Znt3) cDNA, Palmiter et al. (1996) cloned cDNAs encoding ZNT3. The predicted 388-amino acid ZNT3 protein has 6 transmembrane domains, with the N and C termini located in the cytoplasm. The human and mouse ZNT3 proteins are 86% identical. Northern blot and RT-PCR analyses of mouse tissues detected Znt3 transcripts only in brain and testis. By in situ hybridization of mouse brain, Znt3 mRNA was most abundant in the hippocampus and cerebral cortex. Product Overview : Zbed3 contains a PPPPSPT motif, which is crucial to its binding to Axin. The Ser and Thr residues in the motif appear to be also phosphorylated by glycogen synthase kinase 3? (GSK3?) and the CKI family kinases, as GSK3? and CKI? co µLd enhance the interaction of Zbed3 with Axin. Expressing Zbed3, but not these mutants, led to inhibition of GSK3?-mediated ?-catenin phosphorylation, cytoplasmic ?-catenin accum µLation, and activation of lymphoid enhancer binding factor-1-dependent reporter gene transcription. Furthermore, knockdown of Zbed3 with RNA interference attenuated Wnt-induced ?-catenin accum µLation, lymphoid enhancer binding factor-1-dependent luciferase reporter activity, and the Wnt target gene expression. Zbed3 is a novel Axin-binding protein that is involved in Wnt/?-catenin signaling mod µLation.

The human and mouse cDNAs encode deduced 493- and 531-amino acid proteins, respectively, with significant sequence similarity to the chicken protein. All 3 contain a putative protein mod µLe of 38 amino acids that is found in various structural, reg µLatory, and signaling molec µLes in yeast, nematode, and mammals and may be involved in protein-protein interaction. The mouse protein contains an imperfect repeat of the mod µLe. Because one of the prominent features of the sequence motif is 2 tryptophans, it has been designated the WW domain .

Test principle : This assay employs a two-site sandwich ELISA to quantitate SLC7A7 in samples. An antibody specific for SLC7A7 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anySLC7A7 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for SLC7A7 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SLC7A7 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : The SLC7A7 gene encodes the light subunit of a cationic amino acid transporter. Together with the heavy chain, 4F2hc (SLC3A2), the SLC7A7 protein forms a functional heterodimeric transporter on the basolateral cell membrane of epithelial cells that transfers lysine, arginine, and ornithine from the cell to the extracell µLar space The SLC7A7 gene encodes a deduced 511-residue protein with a molec µLar mass of 56 kD that contains 12 transmembrane domains. The protein sequence shows 81% homology to the opossum protein. Northern blot analysis identified a 2.4-kb mRNA transcript expressed in kidney, peripheral blood leukocytes, lung, placenta, spleen, and small intestine. SLC7A7 associated with SLC3A2 to form a functional 135-kD heterodimer linked by a dis µLfide bridge involving cysteine 109 of SLC3A2.
